The Prodigiosin is an lipopolysaccharides drug, which is extracted from cell wall of serratia marcescens. It has a lot of effcacies of anti-inflammatory, antisepsis and anti-infection, at the same time, it can induce emerging of many immune antibodies in order to strengthen the active immunization ability of organism, it is widely used in clinic.But the preparing technology, quality standard and clinical research of it are not well known, so this article uses Serratia marcescens as primitive strain, at the same time, excellent and high product of strains were screened by Ultraviolet mutagenesis, and 16S rDNA gene sequence analysis, physiological and biochemical identification were conducted; fermentation technological parameters were optimized through culturing Serratia marcescens by fermenter, extracts and depurates prodigiosin. At the same time it were researched about pharmacdogy, toxicology and immundogy character istics. The result shows:1.The strains was deal with by Ultraviolet mutagenesis, high product strains was screened when the lethality of the strains suspensions was 70%~80% and the time of exposure was 20-25s. Eight strains was screened firstly by the way of petri dish. Those and primitive strains of SHo was rescreened by shake flask. The biomass of strains body was about 149.3mg/100mL and named as SH-1.2.The result of 16S rDNA gene sequence analysis shows that the similarity of SH-1 and Serratia marcescens strain H3010 16S ribosomal RNA gene (EF194094.1) in database can be 99%. The heterogenicity of Physiological and biochemical identification result between the SH-1 and Serratia marcescens are 99% in the base of SEM.3.It made sure that function of fermenter was stable by inspecting temperature, ventilatory capacity, stirring speed, pH, so it was feasible to fermentation; the best technological parameter of culturing Serratia marcescens with fermenter was temperature of 30℃, ventilatory capacity of 2.5L/min, stirring speed of 160r/min, fermenting time of 72h.4.The crude prodigiosin was made from Serratia marcescens fermented through acetone extraction, mechanical disruption, and deproteinization. The SephadexG-150 purification and HPLC analysis show that the prodigiosin obtained in experiments was accorded with national standard.5.The research of pharmacology and toxicology on mouse indicates that the prodigiosin was safe and reliable; the result of immunology characteristics shows that the prodigiosin has the effects of phagocyte, heighten leucocyte, antibacterial, reinforcing immunization of organism.
