Objective:Comparing the heart shap, function, the cadiocyte apoptosis change and its relationship with 3 kinds of LPA acceptor hypotype of rats after acute myocardial infarction in different time and region, and study the mechanism action of the intervention of promoting blood flow Chinese medicine.Method:To ligate the left coronary artery main stem of rat's heart to make the acute myocardial infarction model. Then divided them into Model, Sham, Xuefu, and PNS groups in random. Picking 48 hours and 5 weeks after the treatment, using Pathological section, haemodynamics measurement, TUNEL and PT-PCR to analyses the functional parameter, cadiocyte apoptosis change and the expression of 3 kinds of lysophosphatidic acceptor gene.Result:Comparing with the model groups, the EF and FS of promoting blood flow medicine group has increased a lot while AI in different zone has obviously decreased (P<0.01).48 hours after MI, the LVSP, +dp/dt max of promoting blood flow medicine group has obviously increased while HB, LVEDP decreased. Comparing with model group, it is statistical significance (P<0.01). Comparing with model group, the expression of LPA1 in promoting blood flow medicine group decreased in statistical significance (P<0.01). Except of the expression of LPA2 in PNS has increased (P<0.01), others has no difference (P>0.05); The expression of LPA3 almost happens in non-infarction and border zone of model group (P<0.05, P<0.01). Medicine group has obvious difference in all zones comparing with model group (P<0.05). MI after 5 weeks, LVEDP has obvious decreased of medicine group, while±dp/dt max increased in statistical significance comparing with model group (P<0.01), especially PNS group (P<0.01). The expression of LPA2 has decreased in statistical significance (P<0.01) comparing with model group in infarction group. For the expression of LPA3, model group increased comparing with sham (P<0.01) in border zone while medicine group has not much difference in non-infarction zone (P<0.05). In border zone, the expression of LPA3 has decreased in statistical significance (P<0.01, P<0.05).Conclusion:The promoting blood flow effect of Chinese medicine can effectively improve haemodynamics value; systole and diastole function after acute myocardial infarction. During different part of time, the Chinese medicine can have depressant effect of myocardial cell apoptosis. In initial stage, LPA 1 and LPA 3 mostly mediate the apoptosis access, the effect happened in marginal zone; the intervention effects of Chinese medicine can active suppression the express of LPA1 and 3. The effect may happen to intervent the access of acceptor for LPA1 and 3 mostly in marginal zone. PNS can have effect of three acceptor of LPA. In the convalescence stage, three acceptor are all join the remodeling process of myocardium, and LPA2 mainly joins the hypertrophic and generation of myocardium. At the same time, LPA1 and 3 mediate the hypertrophic, generation and apoptosis process of myocardium. During the convalescence stage, the acceptors of LPA can be influenced by promoting blood flow medicine for myocardium remodeling mostly in marginal zone. The deducition above should be verificated by cytology.
