Objective To investigate the impact of mPEG on islets survival in homogeneous mice islet transplantation,and find out the impact of PEGylation combined with Rapamycin on anti-rejection therapy in homogeneous mice islet transplantation.Methods The subcutaneously prevascularized STZ-induced diabetic mice treated with transplanted islets of BALB/c mice were randomly assigned into 6 groups:Group A,using normal mice islets; Group B,using the PEGylated islets; Group C,using normal mice islets and administer 1.5mg/kg/d of Rapamycin; Group D,using the PEGylated islets and administer 1.5mg/kg/d of Rapamycin; Group E,using normal mice islets and administer 3mg/kg/d of Rapamycin; Group F,using the PEGylated islets and administer 3mg/kg/d of Rapamycin. Blood glucose after transplantation were monitored,and transplanted islets were analyzed by H&E and insulin immunostain when they were rejected or survived for 6 weeks.Results 1.The survival time in group B was significantly prolonged as compared with group A(P<0.01).2.The survival time in group C were (35.0±3.1)d,and groupD,E,F were survival up to 6 weeks.3.Transplantation sites of group A were observed more severe infiltration of immune cells than group B,and the unmodified islets in group A were completely destroyed after transplantation.4.Insulin-positive islet cells were not detected in the whole area of the transplantation site in group A,while presence in the transplantation site in group B. Conclusions PEGylated islets can significantly improve the survival time of transplanted islets,and combined with Rapamycin could reduce the dose of Rapamycin.
