Objective To evaluate the efficacy of lipid-lowering in vitro therapy for cerebral ischemia and provide a theoretical basis for clinical treatment by the experiments of plasma filtration in vitro in the rabbit model of focal cerebral ischemia.Methods Application of New Zealand white rabbits, high fat diet 5 weeks caused hyperlipemia before the middle cerebral artery occlusion (MCAO) model of cerebral ischemia. Blood was collected with the blood taking bags and centrifugated into blood plasma and blood corpuscle after half an hour of the surgery and reinfusioned in vivo for plasma perfusion treatment after filtered by the filters of plasma lipoproteins. The total of treatment blood is 120ml, The experiment include the treatment group and the control group, each group have 5 rabbits and drawing materials at 1D or 7D. Triphenyltetrazolium chloride (TTC) staining and Hematoxylin-Eosin staining were used to confirm the morphological alteration of brain tissue. ELISA was used to measure the levels of inflammatory factors. Nissl staining was used to observe the damage situation of neuron cell's. Immunity histochemistry was used to observe the survival condition of neurons as well as the activation situation of neuroglia cells. The TUNEL staining was used to observe the apoptosis condition of neurons.Results H-E staining can be observed that the nerve cell's were serious degeneration,necrosis and lossed in the control group, and the degree of injury was significantly reduced in the treatment group; TUNEL staining can be observed the number of apoptotic cells were significantly lower in the treatment group at 7D than the control group (P<0.05); Nissl staining can be observed neurons loss, the outline of neurons was obscure and the structure was unclear in infarct area in the control group; But the neuronic situation was greatly improved in the treatment group, the number of neurons were significantly increased (1D:P<0.05;7D: P<0.01);NSE immunostaining showed that IOD value in the treatment group was significantly higher compared with the control group (1D:P<0.05;7D:P<0.01);GFAP immunostaining also showed that IOD value in the treatment group was significantly higher compared with the control group (1D:P<0.05;7D:P<0.05); CRP levels in brain tissue measured with ELISA in the treatment group at 1D was significantly lower compared with the control group (P<0.05);TNF-a and Ox-LDL levels in brain tissue in the treatment group at 7D was significantly lower compared with the control group (P<0.05)Conclusion Lipid-lowering in vitro therapy can reduce the level of inflammation in rabbits with acute cerebral infarction, promote astrocyte activate and display its functions to protect neurons, decrease neuronal damage and apoptosis, reduce ischemic brain injury.
