| Spleen yang deficient syndrome is a common syndrome in traditional chinese medicine clinic.For yang deficiency result in cold syndrome, stomach yang deficiency always comes with spleen yang deficiency, it also be called spleen and stomach deficiency-cold syndrome.Clinical patients with spleen yang deficiency not only presents clod body and limbs, arms and legs ice-cold, fearing cold and preferring warmth,but also turn up inappetence and tumescence, abdominal pain and prefer pressing, diarrhea with undigested food in the stool, et al. Yang deficiency is related to the hypothermogenesis, spleen deficiency is related to the indigestion. Aconitum Lizhong pill is a classic recipe for clinical treatment of spleen yang deficiency, it has exact effect and few side effect, it's improvement of gastrointestinal symptoms has been researched deeply, but the mechanism of its effect on on hypothermogenesis is still undefined. An experiment was designed to explore this point. Aconitum Lizhong pill was observed in the experiment concerning the regulating efficacy on the models of animals suffering from deficiency of spleen yang.This thesis is divided into literature overview and empirical study.1 OverviewSituation of study of spleen yang deficient syndrome2 Experiment research2.1 Methods of experimentEighteen litters were used in this experiment. Five littermates in each litter whose body weight similarity were bred sequentially and randomized into 5 groups:Normal group,Yang deficiency group, Model group, cold-treated group and warm-treated group. There were 18 rats in each group:Normal group:the rats fed with common feed,and intragastric administration(ig) physiological saline from d70; Yang deficiency group:Thyroidectomy was carried out with 131iodine to destroy partly thyroidal tissue at dl after birth, the interscapular brown adipose tissue was removed at d42, others are same to normal group. Model group:littermates were fed with more cholesterol ester and triglyceride from d56, others are same to yang deficiency group; Cold-treated group:administered intragastrically Huanglian-Jiedu Decoction to inhibit chronic thermogenesis from d70 to d98, others are same to model group; Warm-treated group: administered intragastrically Aconitum Lizhong pill to promote chronic thermogenesis from d70 to d98, others are same to model group. The thermogenesis induced with isoproterenol was evaluated in thermoneutrality (28℃) at d98, phlebotomized from abdominalaorta and extracted sliver and skeletal muscles to measure indicators.2.2 Observation indexAdaptive thermogenesis peak value (℃) and thermogenesis index (℃·h) of the core temperature Body weight and liver indexUncoupling protein 3 in skeletal muscleLevels of serum glucose, triglyceride and total cholesterolMyosin ATPase activity of skeletal muscleEnergy charge in liver and skeletal muscles2.3 The results of experiment2.3.1 The influence of Aconitum Lizhong pill on adaptive thermogenesis in rats with spleen yang deficient syndromeCompared with the normal, the weight growth ratio decreased 16.6%(P<0.01) in Yang deficiency rats, the peak value lowed 5.5%(P<0.01) and thermogenesis index decreased 5.6%(P<0.01). The weight growth ratio of rats treated with Aconitum Lizhong Pill decreased 58.8%(P<0.01) than that in the model rats, the peak value increased 1.1%(P<0.01), and thermogenesis index increased 0.9%(P<0.01).2.3.2 The effect of Aconitum Lizhong pill on uncoupling protein 3 in skeletal muscle of rats with spleen yang deficient syndromeBeing compared with the normal, the UCP3 level in Yang deficiency rats decreased by 7.6% (P=0.439), companied with serum glucose decreased by 24.1%(P<0.01) and triglyceride decreased by 44.7%(P<0.01) and total cholesterol increased by 24.9%(P<0.01). Being compared with the Yang deficiency rats, the UCP3 level in the model rats increased by 22.3%(P<0.01), companied with serum glucose increased by 58.4%(P<0.01), triglyceride decreased by 11.9%(P<0.01) and total cholesterol increased by 33.1%(P<0.01). Being compared with the model rats, the UCP3 level in cold-treated rats increased by 7.7% (P=0.31), companied with serum glucose decreased by 11.7%(P<0.05), triglyceride decreased by 8.2% (P<0.01) and total cholesterol increased by 44.4%(P<0.01); the UCP3 level in warm-treated rats increased by 15.7%(P=0.16), companied with serum glucose decreased by 18.7% (P<0.01), triglyceride decreased by 20.7%(P<0.01) and total cholesterol increased by 9.1% (P<0.01). The UCP3 level in warm-treated rats was 7.4% more (P=0.43) than that in the cold-treated rats. The UCP3 level had a positive correlation with the level of serum glucose (y=0.618x+0.228, R=0.650, P<0.05), a negative correlation with the level of serum triglyceride (y=-0.143x+0.138,R=0.982, P<0.01), and a positive correlation with the level of serum total cholesterol (y=0.649x+0.226, R= 0.722, P<0.01).2.3.3 The impact of Aconitum Lizhong pill on energy charge in skeletal muscles of rats with spleen yang deficient syndromeCompared with the normal, the level of energy charge in skeletal muscle increased by 45.6% (P<0.01) in Yang deficiency rats; the level in model rats decreased 23.9% (P<0.01) than that in Yang deficiency rats; the rats treated with Huanglian-Jiedu Decoction increased the level by 6.8% (P<0.01) and the rats treated with Aconitum Lizhong Pill decreased by 16.8%( P<0.01) than that in the model rats.2.3.4 The impact of Aconitum Lizhong pill on energy charge in liver of rats with spleen yang deficient syndrome Compared with the normal, the level of energy charge in liver increased by 40.8% (P<0.01) in Yang deficiency rats; the level in model rats decreased 23.8%(P<0.01) than that in Yang deficiency rats; the rats treated with Huanglian-Jiedu Decoction increased the level by 9.2% (P<0.01) and the rats treated with Aconitum Lizhong Pill decreased by 22.8% (P<0.01) than that in model rats.2.3.5 The affect of Aconitum Lizhong pill on myosin ATPase activity of skeletal muscle in rats with spleen yang deficient syndromeCompared with the normal, the activity of ATPase in skeletal muscle increased by 101.6% (P<0.01) in Yang deficiency rats; the activity in model rats decreased 20.0% (P<0.01) than that in Yang deficiency rats; the rats treated with Huanglian-Jiedu Decoction increased the activity by 8.0%(P<0.01) and the rats treated with Aconitum Lizhong Pill decreased by 18.6%(P<0.01) than that in model rats.2.4 Conclusion2.4.1 Aconitum Lizhong pill increased the adaptive thermogenesis in rats with spleen yang deficient syndrome.2.4.2 Aconitum Lizhong pill up regulated the UCP3 level in skeletal muscle of rats with spleen yang deficient syndrome. The UCP3 level had a correlation with serum level of glucose, triglyceride and total cholesterol in rats with spleen yang deficient syndrome.2.4.3 Through decreasing the energy charge, at least in part, Aconitum Lizhong Pill was able to enhance significantly the thermogenesis of skeletal muscles in the rats with spleen yang deficient syndrome.2.4.4 Aconitum Lizhong pill down-regulating energy charge in liver of rats with spleen yang deficient syndrome is one mechanism of its promoting thermogenesis potential.2.4.5 Aconitum Lizhong pill down-regulates ATPase activity of skeletal muscle rats with spleen yang deficient syndrome. |
