This paper includes two parts, literature review and experimental study. The literature review includes traditional medicine and modern medicine's understanding of precancerous lesions of experimental breast cancer research, and the Rukang capsules clinical and experimental studies were reviewed; Furthermore, we conduct the experiment to observe the effect of Rukang capsule in rats.Research Objective:To establish rat model of breast precancerous lesions, observe the effects of Rukang capsule on the role of precancerous lesions to explore the role of anti-cancer mechanism of precancerous lesions, to increase Rukang capsule's clinical indications for the use of and provide experimental basis.Methods:90 Female SD rats which were not pregnant, in addition to the control group 15 rats, the remaining 75 rats were initial one-time tube infusion DMBA 100mg/kg body weight, feed every rat 9 weeks to make pathological model of precancerous lesions. After that, the modeling disease model rats were randomly divided into 4 groups, disease model group, tamoxifen group, Rukang capsule low and high dose group. Each group give the scheduled dose for 30 days from the 10 weeks. The ordinary pathology methods, radioimmunoassay, immunohistochemistry method, computer image analysis techniques to observe the general conditions of rats, body weight, height and diameter of the nipple, breast tissue morphology, serum sex hormone E2, P levels, breast tissue VEGF, bFGF, MVD, ER, PR expression, and the results were statistically analyzed.Results:Rukang capsule low and high dose group rats inhibited breast enlargement, nipple diameter and height were significantly different with disease model group (P<0.05-0.001), compared with the tamoxifen group showed no significant difference (P>0.05), while the low-dose effects of Rukang capsule slightly weaker the high dose group, indicating Rukang capsules inhibit breast enlargement in the rat dose-effect relationship exists. Optical microscope to observe the patterns of rat mammary gland found that, DMBA rat after modeling, the performance of the model group is severe pathological features of atypical hyperplasia, showing that the DMBA can well copy precancerous pathological model, the Rukang capsules low and high dose groups and the tamoxifen group performance are simple hyperplasia and mild dysplasia of the main pathological features. Indicate that Rukang capsule and tamoxifen can inhibit and reverse the breast cancer development and progression of precancerous lesions, while Rukang capsule dose-effect relationship exists, the effect of high dose is better than low dose group. Rukang capsule low and high dose groups could reduce the breast precancerous lesions of the content of serum E2, significantly different with the disease model group (P<0.001), and with tamoxifen group was no significant difference(P>0.05); while the content of serum P not significant, compared with the model group was not significant (P>0.05). Rukang capsule low and high dose groups could cut breast cancer ER-positive area and mean optical density, and the disease model group was significantly different (P<0.05-0.001), with no statistical difference between the tamoxifen group significance (P> 0.05), shows Rukang capsule to reduce the high expression of ER; Rukang capsule high dose group may cut breast precancerous lesions of PR-positive area and optical density, compared with the model group significantly different (P<0.05), while the Rukang capsule low dose group and model group showed no significant difference (P>0.05), shows Rukang capsule's effect on reduce of PR expression is obvious, dose-effect relationship between the high-dose effect is obvious, but low-dose effects not obvious. On the result of VEGF, bFGF and MVD:disease model rats exist VEGF, bFGF and MVD's high expression, compared with the control group is significant difference (P<0.001), Rukang capsule low and high dose groups could reduce the the positive area of VEGF, bFGF and MVD, reducing the average optical density of bFGF, compared with the model group were significant differences (P<0.05-0.001), and showed some dose-effect relationship, Rukang capsule high dose group better than the low dose group; while VEGF and MVD, the average optical density not significant, compared with the model group showed no significant difference (P>0.05). Explain that Rukang capsule plays good role on reduced the expression of bFGF, and may be the main ways in inhibiting the angiogenesis, and for no obvious effect on VEGF and MVD.Conclusion:1. Rukang capsule significantly inhibited rats'breast enlargement, reduced the incidence of precancerous breast lesion in rats.2. Rukang capsule reduces the incidence of precancerous lesions in rats is closely related with the mechanism of reducing the serum E2 levels, reduced high expression of ER.3. Rukang capsule reduces the incidence of precancerous lesions in rats is related with the mechanism of reduce the expression of bFGF, and maybe the main way to inhibition of angiogenesis.
