CDK11p58, a CDK11/PITSLRE protein kinase family member, is encoded by Cdc2L2, and it plays a crucial role in cell cycle regulation. Pancreatic cancer, featured by rapid progression and bad prognosis, is a common malignant tumor of digestive system, and pancreatic ductal adenocarcinoma is of the major histological form. The incidence of pancreatic cancer increased rapidly in recent years.It is now unclear about the relation between CDK11p58 and human pancreatic cancer. In this study, we focused our research on the relation between CDK11p58 and the proliferation of human pancreatic ductal adenocarcinoma cell as well as the molecular mechanism.We established a CDK11p58 prokaryotic expression vector, and we screened a cell clone which stably overexpresses CDK11p58 by initially introducing the recombinant eukaryotic expressional plasmid pcDNA3.0-HA-p58 into Human pancreatic ductal adenocarcinoma cell line MIAPaCa-2 via Lipofectamine2000 and successively selected by G418 and finally identified by western blot. Cell cycle, cell proliferation and G1/S related gene mRNA levels were analyzed by flow cytometry assay, MTT assay and real-time PCR assay, respectively.Result showed that the S phase cell proportion, the proliferation activity and the mRNA level of cyclin D1, cyclin D3, p21 gene in pcDNA3.0-HA-p58 transfected cell were significantly increased compare with vector group and control group(p<0.01). These result indicated that overexpression of CDK11p58 can accelerate MIAPaCa-2 cell G1/S progression and promote its proliferation by up-regulating the mRNA level of cyclin D1, cyclin D3 and p21gene. These results provided an important basis to further explore the molecule mechanism of proliferation of human pancreatic ductal adenocarcinoma cell induced by CDK11p58 in vivo and the cure of the disease.
