Objective:Asymmetric dimethylarginine (ADMA), an intensive endogenous nitric oxide synthase (NOS) inhibitor, is thought to be a novel risk factor of cardiovascular diseases.Monocytes/macrophages and foam cells which are associated with macrophages play a crucial role in the development and progression of atherosclerosis.The aim of this study was to investigate the effect of ADMA on cholesterol content in cultured human THP-1 monocyte-derived macrophages and foam cells.Methrods:The human THP-1 was maintained in vitro,differentiation of cultured THP-1 cells into macrophages was stimulated with phorbol 12-myristate 13-acetate (PMA), histostainTM assay was used to assess THP-1 derived macrophages;THP-1 derived macrophages were induced to foam cells by ox-LDL, cellular lipid accumulation was determined by oil Red 0 staining. THP-1 derived macrophages/foam cells were incubated with ADMA of varing concentration (3.75, 7.5,15,30μmol/L) for various periods(6,12,24h),enzyme-linked colorimetry assay were used to determine intracellular cholesterol content.Results:HistostainTM assay results showed that CD68 expression was activated in 85% THP-1 monocytes after incubated with PMA, The THP-1 cell were differentiated into macrophages.Oil Red 0 staining results showed that Intracellar lipid droplets increased, the macrophages were differentiated into foam cells by ox-LDL. Enzyme-linked colorimetry assay results showed that ADMA could increase THP-1 derived macrophages/foam cells cholesterol content in a concentration-time dependent fashion, when the concentration of ADMA is 30μmol/L,the activatory effects was elevated obviously at 24h.Conclusions:1.With concentration-time dependent fashion,ADMA could increase human THP-1 derived macrophages the ability of lipid accumulation and intracellular cholesterol content.2.ADMA could cause typical histopathological hallmark of human THP-1 derived foam cells.increase cholesterol content of human THP-1 derived foam cells,induced intracellular cholesterol accumulation,increase foam-cell formation.3.ADMA may be an important factor contributing to the formation of THP-1 derived macrophages and foam cells, might be new drug targets for the clinical study of arteriosclerosis.
