Purpose:Our study aimed to detect the change of tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β)in serum and hypothalamus of endotoxin-induced fever rabbits after administration and to discuss the effect of coptis decoction on the antipyretic effect and mechanisms of endotoxin-induced fever rabbits.Methods.Sixty general-level New Zealand rabbits, male, weighted 2.0±0.5kg, were housed individually in a room and free of water and food, feed in ventilated lab at 20℃.Rabbits were randomly divided into four groups:saline group; endotoxin group;coptis decoction plus endotoxin group;ibuprofen plus endotoxin group.Endotoxin was injected through intravenous administration.Saline, coptis decoction and endotoxin were through gastrointestinal administration. We observed the temperature change from 0 to 5 hours, and conducted the statistical analysis based on the detected content. Results:Coptis decoction has remarkable antipyretic effect on the fever rabbits. Moreover, the contents of TNF-α,IL-1βin the blood and hypothalamus of coptis decoction plus endotoxin group were lower than those in the endotoxin group.1.Coptis decoction can reduce the body temperature of endotoxin-induced fever rabbits, and two-phase hot disappeared.2.Coptis decoction can decrease the contents of IL-1βand TNF-αin the blood and hypothalamus of fever rabbits induced by endotoxin and inhibit the temperature elevation. Conclusions:Coptis decoction has remarkable antipyretic effect on fever rabbits induced by endotoxin, its mechanism may be related to the conditionthat this formula can reduce the content of TNF-α,IL-1βin the blood and hypothalamus of fever rabbits and further inhibit temperature elevation.
