Purpose:The object of this thesis was found that the difference of protein in yuan-point and zangfu of rat kidney through the method of two-dimensional electrophoresis and mass-spectrometric technique,and investigate the homogenous of protein in yuan-point and zangfu.Material and methods:12 Wistar rats were divided randomly into blank control group(n=6) and "taixi" group(n=6)."taixi" group was treated 7 times by electroacupuncture at "taixi" point and after treatment,the total proteomes of kidney,the tissue of "taixi" point and non-acupoint were compaired and analysised by two-dimensional gel electrophoresis and image analysis software(ImageMasterTM2D Platinum electrophoresis image analysis).The related proteins were analyzed by matrix-assisted laser desorption ionization - time of flight mass spectrometry(MALDI-TOF-MS).Results:1.Matching the non-acupuncture point and the "Taixi" point,the tissue of "Taixi" point contained the 8-point difference.2."Taixi" point showed significant changes of proteome afer acupuncture.3.non-acupoint and kidney showed 10 related proteins,but it was less than "Taixi" Point4."Taixi" Point and kidney was detected 13 related proteins,the results showed that "Taixi" Point have more related protein with kidney than non-acupoint.5."Taixi" Point showed 20 related protein with kidney tissue after acupuncture.6.After acupuncture,9 proteins showed 3-fold difference in kidney.7.NAD-dependent isocitrate dehydrogenase(NAD-IDH) was found "Taixi" point and kidney after mass spectrometry analysis.Conclusion:The protemes of non-acupuncture point and "Taixi" point showed significant difference,but non-acupuncture point and "Taixi" point contain the same protein related with kidney."Taixi" point showed more proteins related with kidney than non-acupuncture point.After acupuncture at "Taixi" point,the proteme of "Taixi" point tissue showed significant difference,and we found more proteins related with kidney than preacupuncture.In kidney,9 proteins showed 3 fold increase after acupuncture,and these proteins were analyzed with MALD-TOP/MS.Rinally the results were analyzed at NCB1 database.We found a interesting protein NAD-IDH.We considered that acupuncture stimulated and activated some fountional proteins in acupuncture point and tissue and NAD-IDH is one of the effective materials when acupuncture at "Taixi" point.
