Purpose:1.To optimize the processing technology of Rhizoma Alismatis,based on chemical constitents change.2.To illustrate processing principle of Rhizoma Alismatis,based on toxicity and pharmacodynamics change.3.To establish the quality standard of Rhizoma Alismatis,Rhizoma Alismatis with salt water and Rhizoma Alismatis with wheat bran.Material and method:Material:Rhizoma Alismatis was purchased from the Chinese National medical in Bozhou city,Anhui province.A voucher specimen was deposited at our laboratory.Method:1.Processing technology research of Rhizoma AlismatisThe sample was prepared with ultrasonic extraction method.The content was determined by HPLC,using 24-Acetyl alisol A and 23-Acetyl alisol B as index, establish the optimum processing technology.2.Toxicity and pharmacodynamics study of Rhizoma Alismatis and its processed products.Employ the maximal experiment does to examine the acute toxicity of Rhizoma Alismatis.Through weighing ear edema and the immune organs of rats,to examine the anti-inflammatory and anti immune actions of Rhizoma Alismatis.3.Quality standard research of Rhizoma AlismatisWe establish the quality standard of Rhizoma Alismatis and its processed products,through testing moisture content,heave metal limit examine,total ash examine and total extractives limit examine,etc.4.Inorganic elements determined of Rhizoma Alismatis and its processed productsWe adopted ICP-MS method to determine the inorganic elements of Rhizoma Alismatis and the processed products with.Results:1.Processing technology research of Rhizoma Alismatis Via single factor experiment and L9(34) orthogonal design,we obtain the best processing technology of Rhizoma Alismatis.researched the stir-bake with saltwater technology on Rhizoma Alismatis and processing of stir-frying with bran for Rhizoma Alismatis with HPLC.The optimum stir-bake with saltwater technology was:Rhizoma Alismatis mixed with 2%salt water thoroughly,sealed moisting,stir-frying for 35 minutes,110℃for pot tempreture.The optimum stir-frying with bran technology was:Rhizoma Alismatis adding amount of 10% bran mixed thoroughly,tir-frying for 3 minutes,170℃for pot tempreture.2.Toxicity and pharmacodynamics study of Rhizoma Alismatis and its processed products.Through acute toxicity experiment on rat,we observed that the Rhizoma Alismatis and the processed products were safe on clinical use.Via weighing the ear edema and the immune organs of rats,we observed that the anti-inflammatory and anti immune actions of the Rhizoma Alismatis and the processed products were strengthened.3.Quality standard research of Rhizoma AlismatisEstablish the quality standard of Rhizoma Alismatis,Rhizoma Alismatis and Rhizoma Alismatis with wheat bran.The content of 24-Acetyl alisol A and the 23-Acetyl alisol B is ont less than 0.015%and 0.06%in Rhizoma Alismatis.The content of 24-Acetyl alisol A and the 23-Acetyl alisol B is not less than 0.025% and 0.05%in Rhizoma Alismatis with salt water.The content of 24-Acetyl alisol A and the 23-Acetyl alisol B is not less than 0.03%and 0.075%in Rhizoma Alismatis with wheat bran.4.Inorganic elements determined of Rhizoma Alismatis and its processed products Through determination on Rhizoma Alismatis and its processed products of inorganic elements with ICP-MS,the content of inorganic elements was high in Rhizoma Alismatis and its processed products.In the content of inorganic elements in processed products has not changed.The content of heavy metal elements was in accord with Pharmacopoeia standard. Conclusion:1.Optimum processing technology of Rhizoma Alismatis was obtained,the test of magnifying experiment was consistent with lab experiment.The technology was stable and available.2.Through acute toxicity experiment on rat,we observed that the Rhizoma Alismatis and the processed products were safe on clinical use.Via weighing the ear edema and the immune organs of rats,we observed that the anti-inflammatory and anti immune actions of the Rhizoma Alismatis were more strengthened than the processed products3.Establish the quality standard of Rhizoma Alismatis,Rhizoma Alismatis and Rhizoma Alismatis with wheat bran,through testing moisture content,heave metal limit examine,total ash examine and total extractives limit examine,etc.The quality standard is stable and feasible.4.From ICP-MS experiment,we concluded the content of inorganic elements was high in Rhizoma Alismatis and its processed products,and trace elements has no change.The heavy metal elements was in accord with Pharmacopoeia standard.
