Objected: To construct the fusion gene Hyper-IL-6, and the recombinant lentiviral vectors encoding Hyper-IL-6. Detect the viral titers of the viral particle and the expression of Hyper-IL-6 in the human hepatoma cell lines HepG2 treated with FIV-HIL-6.Methods: Using GeneSOEing method, a cassette consisting of human sIL-6R DNA and IL-6 DNA was fused by a linker coding for sequesnes rich in glycine by PCRtechnique and cloned into pCEDF. The plasmid pCDEF-Hyper-IL-6 and the other two packaging plasmid were co-transfected into 293T cells. The viral supernatant was collected. The viral titer was detected and the expression of usion gene encoding Hyper-IL-6 was detected in the HepG2 cell lines treated with FIV-Hyper-IL-6.Results: The recombinant lentiviral vectors encoding Hyper-IL-6 was successfully constructed. The titer of recombinant lentiviral virus and the expression of the fusion gene in the HepG2 cell lines were detected successfully. Conclusion: The success in construction of the lentiviral vectors encoding Hyper-IL-6 and the expression of the fusion gene in the HepG2 cell lines make it possible to study further on the continued research. Objected:To study the stimulation of Hyper-IL-6 to L-02 cell lines in expression of Haptoglobin, and investigate the promotion of Hyper-IL-6 to L-02 cells in generation.Methods: The expression of Haptoglobin in L-02 cell lines was analysised with RT-PCR techniques after being treated with FIV-Hyper-IL-6. The growth rate of L-02 cells was analysised with MTT techniques at different time point after being treated with FIV-Hyper-IL-6. Results: RT-PCR test indicated that the expression of Haptolobin in L-02 cells treated with FIV-Hyper-IL-6 was increased obviously (F=7.85). The data of MTT analysis showed that both Hyper-IL-6 and IL-6 can promote L-02 cells to promote, and the promotion of Hyper-IL-6 was much stronger than IL-6 (p=0.0000).Conclusion: Compare to IL-6, Hyper-IL-6 has the more conspicuous biological activity to intimate the expression of Haptolobin and the growth of the L-02 cell lines.
