Differential Protein Expression Of Different Prevention And Treatment Of Acupuncture Acupoints Stress Ulcer In Rats

Purpose:The paper was on basis of the findings in researching the effect rule and mechanism of SU by"He-Mu conbination"on the stomach meridian, and IEF/SDS-PAGE was applied in the experiment for investigation into material foundation of acupuncture effect. This paper was made to provide the basis to the acupuncture compatibility and acupoint collabrative study.Method:96 Wistar rats were divided into eight groups stochastically: blank group, colligation group, model group,"Zusanli"group,"Zhongwan"group,"He-Mu point conbination"group,"Shu-Mu point combination"group, eight confluent point group. The gastric ulcer rats model were copied by colligation and water immersion. Each group of rats were acupunctured according to the grouping,"Zusanli"group was taken"Zusanli"(ST36) acupoint;"Zhongwan"group was taken"Zhongwan"(RN12) acupoint;"He-Mu point conbination"group was taken"Zusanli"(ST36) and"Zhongwan"(RN12) ;"Shu-Mu point combination"group was taken"Zhongwan"(RN12) and"Weishu"(BL21) accupoint; eight confluent point group was taken"Neiguan"(PC6) and"Gongsun"(SP4) accupoint; model group was colligated every day. Immerce 6 groups in the thermostatic waterbath after two days; colligation group was only colligated everyday, blank group was fed without handling. All the stomach of rats were analysesd by IEF/SDS-PAGE and compared with each other.Results:1 Comparing visual observation of colligation group with model group, the former one have some ulcer focuses whose quantity and size are not equal; the degree of gastric mucosa injury in acupuncture treatment group is lighter than model group.There are some bleeding points in"Zhongwan"group,"Zusanli"group,"Shu-Mu combination"group, and eight confluent group, but the differences are not obvious."He-Mu combination"group has a few bleeding points but the diameter and amount are very small.2 There are significant differences in colligation group, model group,"Zhongwan"group,"Shu-Mu combination"group, eight confluent group and"Zusanli"group in the ulcer index. There are very significant differences between model group and"He-Mu"group. There are less significant differences between"Zhongwan"group,"Zusanli"group,"Shu-Mu combination"group and eight confluent group in the ulcer index. There are significant differences in"Zhongwan"group,"Zusanli"group,"Shu-Mu combination"group, eight confluent group, and"He-Mu"group in the ulcer index.3 There are different protein points of model group, colligation group and acupuncture treatment group(except"He-Mu conbination"group) to the blank group in different levels. But the protein map of"He-Mu conbinatiln"group has no signifecant differences with normal group.Conclusion:1 Acupuncturing"Zusanli"(ST36) point or "Zhongwan"(RN12) point both can play a role in gastric ulcer model, but eighter one is inferior to using therm at the same time."He-Mu conbination"group has differences in protain expresion patterns with other point conbining group. It shows that He-Mu conbination must have the effect of specific.2 After made the stress gstric ulcer modle, the protain expressions profile in rat gastric tissu were changed. Acupuncture treatment has a ragulatory role in rat gastric ulcer tissu protain expression., and so dose He-Mu conbination treatment. Furthermore, the simple between pure bundle stress bundle ad imersion stress also has signifficant diferfrence, and it provides basis to experimental animal modle copy.3 Accupuncture effect to the change of proteome is holistic, bidirectional, dynamic, rather than single,unidirectional,stationary.
