Objective:Evidence shows that the morbidity of metablic syndrome is increased quickly in our country,and metabolic syndrome will bring on cardiovascular disease.The morbidity and mortality of cardiovascular disease also enhance fast.Oxidative stress may play a role in the pathophysiology of cardiovascular disease,but little is known about antioxitant status among individuals with the metabolic syndrome who are at high risk for developing these conditions.This study aimed to investigate the association between antioxidants and the clinical biochemistry index.Methods:1.A HPLC method for determination of vitamine A,E,lycopene andβ-carotene in serum was established with retinyl acetate as intern standard.Hypersil-ODS column was used.The mobile phase consisted of mobile A:methanol:acetonitrile:distilled water(V:V:V)=6:2:2,mobile B:aceonitrile:methanol:tetrahydrofuran(V:V:V)=5:2.5:2.0.The gradient elution programe of the mobile phase was as table 1,the flow rate was 1.0ml/min,injection 20μl and column temperature 45℃.The wavelength of detection vitamin A,retinyl acetate,vitamin E,β-carotene and lycopene were 325nm,325nm,292nm,450nm and 472nm respectively.2.The serum antioxidants lever and the concentration of clinical biochemistry index in patients with metabolish syndrome was investigated.The concentration of fasting serum glucose,triglyceride, total cholesterold,serum vitamin A,E,lycopene andβ-carotene in 60 patient with metabolic syndrome and 60 healthy subjects were measured. Resultls:1.The average retention time for vitamin A,E,β-carotene and lycopene were 3.7,4.2,6.8,8.3 and 10.5min.The recovery of vitamin A was from 91.0% to 95.8%.The recovery of vitamin E was from 98.0%to 86.4%,The recovery of P-carotene was from 105.0%to 97.6%.The recovery of Lycopene A was from 117.0%to 106.4%.The intra-day RSD of vitamin A,E,β-carotene and lycopene were 3.47%,3.94%,17.91%and 5.80%,the inter-day RSD of vitamin A,E,β-carotene and lycopene were 3.54%,3.89%,2.28%and 7.78% respectively.The standard curve of vitamin A was linear in the range of 0.14—7μg/ml.The standard curve of vitamin E was linear in the range of 2—100μg/ml.The standard curve of P-carotene and lycopene were linear in the range of 0.04—2μg/ml.In 60 normal residents,the mean serum concentration of vitamin A,E,β-carotene and lycopene were 0.624±0.254μg/ml,12.095±4.125μg/ml,0.188±0.085μg/ml,0.378±0.244μg/ml respectively.1.Compared with the healthy subjects,the average value of Pcarotene (0.282±0.193μg/ml) in MS group was lower(P<0.05),the concentration of serum triglyceride(2.29±1.37 mmol/L) was higher (P<0.01).R value of Pearson correlation coefficients betweenβ-carotene and triglyceride was negative(P<0.01).The result of Multiple linear regression analysis showed that after adjusted by sex and age,β-carotene was negative correlated with triglyceride.Conclusion:The HPLC method for determination of vitamin A,E,lycopene andβ-carotene in serum was accurate.Sample handling was relative quick and simple.There was a close relationship between the serum antioxidants and the clinical biochemistry index in patients with metabolic syndrome, The concentration of triglyceride would be decerased along with the lever of serumβ-carotene increased.
