Objective To study the effect of adenovirus-mediated tandem expression of two RhoA and two RhoC siRNA on the growth of human colorectal carcinoma transplanted in nude mice.Methods Human co lorectal carcinoma HCT116 cells were anabiosissed,cultivated and passaged enough.And then HCT116 cells were implanted subcutaneously into nude mice.The model of transplanted colorectal HCT116 cell line of colorectal carcinoma in nude mice was set up.Twenty one tumorigenesis nude mice were separated into three groups randomly:The first group is adenovirus vector with RhoA and RhoC in tandem expression group(Ad-RhoAC);the second group is adenovirus vector with plasmid of HK group(Ad-HK);the third group is normal sodium group(NS).The given dose corresponding interferent(The Ad-RhoAC group was injected adenovirus vector with RhoA and RhoC in tandem expression;the Ad-HK group was injected adenovirus vector with plasmid of HK;the NS group was injected normal sodium.) was injected into each mouse.The transplanted colorectal carcinoma volume and weight were observed.In order to find out the difference,we analyzed the data of the transplanted colorectal carcinoma volume,weight,growth rate and inhibition rate.Results The mean volume of Ad-RhoAC group,Ad-HK group and NS group was (441.38±63.03) mm~3,(678.81±155.39) mm~3 and(699.62±190.56) mm~3;the mean growth rate was(2.38±0.56),(4.58±0.94) and(5.05±0.48);the mean weight was (0.36±0.13) g,(0.78±0.22) g and(0.75±0.22) g,respectively.The Ad-RhoAC group was different from Ad-HK group and NS group in the mean volume,the mean growth rate and the mean weight(P<0.05).There was no significant difference between Ad-HK group and NS group in the mean volume,the mean growth rate and the mean weight(P>0.05).The growth of transplanted colorectal carcinoma was significantly inhibited in the Ad-RhoAC group,and the inhibition rate was 44.38%.The inhibition rate of the Ad-RhoAC group was higher than that of the Ad-HK group and the NS group.Conclusion Adenovirus-mediated tandem expression of two RhoA and two RhoC siRNA can obviously inhibit the growth of human colorectal carcinoma transplanted in nude mice.
