Objective To Investigate the expression of Heme Oxygenase-l(HO-1) and influences on Retinal Ganglion Cells(RGCs) following animal models of acute intraocular hypertension in rat retina and the intervention of Huoxuehuayu Decocti in order to study the effect of Huoxuehuayu Decocti on glaucomatous acute retinal injury.Methods 120 Wistar rats were devided into 4 groups at random,including group A,B,C and D.Group A was used for Real-Time quantitative reverse transcription-polymerase chain reaction(Real-Time qRT-PCR).Group B was for hematoxylin and eosin stain(HE stain).Group C was used for transmission electron microscope(TEM).Group D was for immunofluorescence staining(IF).Each Group was devided into 10 small groups,and all left eyes were taken.Take group A for example,A0 were normal control group,A' were punctura group,A1~A4 were intraocular hypertension group,A5~A8 were intraocular hypertension involved Huoxuehuayu Decocti group.A',A1~A8 group was modeled by increasing anterier chamber pressure.The retinas of A1~A4 group were taken out at 1d, 3d,7d,14d after intraocular hypertension.Rats of A5~A8 group were given Huoxuehuayu Decocti right after intraocular hypertension,and the retinas were taken out after feeding the medcine of 1d,3d,7d,14d.The small groups of other Group were similar.Results 1.Real-Time qRT-PCR:Expression of HO-1mRNA was upregulated and peaked 1 day after intraocular hypertension.The expression of intraocular hypertension involved Huoxuehuayu Decocti groups were higher than intraocular hypertension groups at the same time points.2.Light microscope:At the early stage,the inner layers of retina had edema,and at the advanced stage the number of RGCs decreased and the thickness of retinal nerve fiber layer were thinner.The damage were lighter in intraocular hypertension involved Huoxuehuayu Decocti groups compared with intraocular hypertension groups at the same time point.3.TEM:1 day,3 days after intraocular hypertension,the mitochondria, endoplasmic reticulum in ganglion cells and nerve fibers became swelling.Apoptotic bodys could be seen in the RGCs layer 7days,14days after intraocular hypertension.The number of apoptotic RGCs in in intraocular hypertension involved Huoxuehuayu Decocti groups was lower than that of intraocular hypertension groups.4.Immunofluorescence staining revealed that HO-1 expressed in retinal outer nuclear layer,inner nuclear layer and inner plexiform layer after intraocular hypertension.Conclusion Rat retinal ischemia-reperfusion injury caused by acute intraocular hypertension could induce the expression of HO-1.According to its function of anti-oxidative stress,HO-1 could reduce the damage and apoptosis of RGCs caused by acute retinal damage.Huoxuehuayu Decocti could upregulate the expression of HO-1, and it might play a part in preservation by enhancing the function of HO-1 during retinal oxidative stress.
