Mesenchymal stem cells(MSCs) are highly self-renew and multipotent adult stem cells from mesoderm. It can be isolated from many connective tissues and mesenchyma of some organs. It becomes a kind of perfect seed cells of tissue engineering because it has abilities to differentiated into multiple lineages and expand in vitro easily. Placenta and umbilical cord belong to fetal appendage,which are discarded after parturition. However, part of them derived from extraembryonic mesoderm. If MSCs could be obtained from placenta and umbilical cord,we will have a large seed cell bank for tissue engineering.Part one: we isolated MSCs from placenta and umbilical cord, and cultured in vitro. Then compared them with MSCs from bone marrow in shape, character, surface marker, and diferentiation abilities. Furthermore,we induced MSCs of above three resources to differentiate into adipocytes,ossified and endothelial cells. The result demonstrated that we isolated stable MSCs on cell morphology and biological characteristic successfully. Three kind of MSCs had no difference in shape, surface marker, and diferentiation abilities. But MSCs from bone marrow had stronger immunogenicity and longer culture doubling time than the other kinds of MSCs.Part two:we have done researches on the application of MSCs from placenta in tissue engineering. In the first part, we found that MSCs from Placenta have a characteristic as follows:easy to get,high successful rate to isolate and culture, and stronger expanding ability. So we selected MSCs from Placenta as seeds of tissue engineering. Transplant MSCs from Placenta and MSC-EC on Gel/MMT-CS surface,cell culture flask as a control. The result demonstrated that both MSCs and MSC-EC can survive and proliferate on Gel/MMT-CS surface. There is no difference between MSCs and MSC-EC. But control(cell culture flask) has obvious difference.Part three: we took some studies about differential RNA display after MSCs diferentiated into vascular endothelial cells. Throngh gene-chip as a high-flux filter to find differential RNA display, we analysed gene expressing changes of MSCs in the process of diferentiating into vascular endothelial cells. The results demonstrated that there were many changes of genes encoding several adhensin molecule and ECM and vascular endothelial cells function-related proteins and receptors in the process of diferentiation into vascular endothelial cells.To sum up, we first brought to success to isolate and culture MSCs from placenta, and explored biological characteristic, which established the foundation of further studies.Also,we explored the primary application of placenta MSCs in tissue engining and the gene expressing changes in the process of differentiating into vascular endothelial cells, that provided a basis to the future application and research of MSCs.
