The treatment of bronchopleural fistula(BPF) with medicine submucosal injection through a flexible bronchoscope on animal modelsPurpose To study the expressions of vascular endothelial growth factor(VEGF) and smooth muscle actin(SMA) in SD Rats' trachea submucosa and to observe the capillary and fibroblast with microscopes and to observe if cardiografin,tetracycline can take the place of polidocanal.Methods 102 SD Rats were divided into normal(6 Rats),normal sodium injection (24 Rats) group,polidocanol injection(24 Rats) group,cardiografin injection(24 Rats) group,tetracycline(24 Rats) group.The latter four groups were equally divided into four subgroups according to the time of 1 st day,3rd day,5th day and 7th day.The Rats' trachea submucosa were injected with 0.05ml normal sodium or polidocanol or cardiografin or tetracycline,and the normal group without any interference.All the samples were stained by hematoxylin-eosin(HE) or immunohistochemistry,and were observed by light and electron microscopes.And the positive area and integrated optical density(IOD) of VEGF and SMA were analyzed with Image Pro Plus 5.1 software.Results 1.According to the light microscope:The capillary of trachea submucosa with normal sodium injection were more than the normal,but were much less than those in the polidocanol injection group,cardiografin injection group or tetracycline injection group.2.According to the electron microscope:The fibroblast in the polidocanol injection group,cardiografin injection group and tetracycline injection group were much more than those in the normal.Collagen fiber bundles were present and became denser and more tightly packed.3.Polidocanol,cardiografin,tetracycline can distinctly increase the expressions of VEGF and SMA in SD Rats' submucosa and visibly promote the new capillary and fibroblast formations.4.Within the polidocanol injection group,cardiografin injection group and tetracycline injection group,the effect was much more obviously in the 3rd day than in the 1st day(P<0.01), and it was much more obviously in the 5th day than in the 7th day(P<0.01),the apparently reaction time was from the 3th day to the 5th day..5.Among these three groups,in the 3rd day there were no difference among them(P>0.05).In the 5th and 7th day it was much lower in the polidocanol injection group than that in the cardiografin injection group or tetracycline injection group(P<0.01). Conclusions 1.Cardiografin,tetracycline can take the place of polidocanal.2. Cardiografin,tetracycline can be used in the clinical treatment for further study.
