Objective:To investigate the effect of heme oxygenase 1(HO-1) on the expression of tumor necrosis factor-alpha(TNF-α) in RAW264.7 cells,and investigate the mechanism of antiatherosclerosis of heme oxygenase 1Method:The foam cells were established by the cultured RAW264.7 cells(mouse macrophage) that were incubated with oxidized LDL.Hemin and ZnPPIX were used to effect on foam cells to express heme oxygenase 1 and immunocytochemistry method were used to test the effect of HO-1 on expression of tumor necrosis factor-alpha.Results:(1) HO-1 detectation:HO-1 weakly positive cells were observed in control group in RAW264.7 cells.The expression of HO-1 in foam cells was increased. Induced HO-1 by Hemin that used in different experiment concentration(5, 10,50uM) was profoundly increased and dose-dependent.ZnPPIX(10 uM) group was decreased.The difference of HO-1 expression compared with control groups is significant(p<0.05).(2) TNF-αdetectation:TNF-αweakly positive cell were observed in control group in RAW264.7 cells.The expression of TNF-αin foam cells was increased. The expression of TNF-αin different Hemin concentration(5,10,50uM) was profoundly decreased and dose-dependent.ZnPPIX(10 uM) group was increased. The difference of TNF-αexpression compared with control groups is significant(p<0.05).Conclusion:Induced HO-1 by Hemin can decreased the expression of TNF-αand decreased HO-1 by ZnPPIX can increased TNF-a expression in RAW 264.7.It demonstrated HO-1 can inhibit TNF-a,which may be one of the mechanism of anti-atherosclerosis of HO-1.
