Jatamansi Extract Process And Dosage Forms

This paper focuses on Valeriana Jatamansi J.(Zhi Zhuxiang)and its effects on relieving the symptom of IBS(Irritable bowel syndrome).By means of modern chemistry techniques such as extraction,seperation and shaping,iridoids,one of Zhi Zhuxiang's major components,is easy,because of its natural instability,to degrade into Homobaldrinal,which is the final and actual component to relieve the IBS disease.The main research process and significant results are listed as follows:(1) Optimazation of extraction technology and determination of chemical compositions of Homobaldrinal,again coincident with standards of Chinese pharmacopoeia 2005.(2) Identification of chemical components of Homobaldrinal and a chemical reference substance preparation.The reference substance purity is more than 98.07%, coincident with the standard of determination of chemical compositions.(3) By means of orthogonal design,the best extraction requirements are corresponding pharmacy passing through 24 mesh sieve,to hot reflux with 12 times of 70%ethanol,extracting for 3 hours and 2 times in all.By AB-8 macroporous resin, maximum adsorption is 22.6mg/ml,concentration of upper sample is 1.42mg/ml, adsorption flow rate is 2BV/h,elution solvent is 90%ethanol,elution flow rate is 2BV/h,and to 5 times volume.(4) Option of oral dose.The therapeutic effect and the mechanism reseach of Zhizhuxiang on IBS.Firstly screening the pharmacological active site of Zhizhuxiang on the gastrointestinal motility.Seconedly,screening the pharmacological active site of Zhizhuxiang on the neuropsychic fuction.After pharmacological experiments, proposed preliminarily oral dose is 18mg.(5) Preparation of Valeriana Jatamansi J.sustained-release matrix tablets.Some apposite technology parameters of the tablet are established.According to the technology parameters,I filtrated the best framework material of the sustained release matrix tabletand studied the Pharmaceutical technology of it.The Result:EC and HPMC is the perfect framework material,the matrix tablet made by them coincide to thetechnology parameters and can achieve ideal sustained result The best prescription ration is Zhizhuxiang extract:HPMC:EC:lactose:magnesium stearate=85:150:15:48.5:1.5.(6) To estabilish qualify standards of Valeriana Jatamansi J.sustained-release matrix tablets.The Valeriana Jatamansi J.in tablet was identified by using TLC;the content of the matrix tablet was determined by using HPLC and the content limit was established;the release test methods and result were established.Result:The quality standard was established preliminarily.This method is stability,accurate,specific and repeatable.It can control the quality of the matrix tablet well.These standards are helpful to control the qualify of the drugs when manufacturing.(7) This paper does comprehensive research on Homobaldrinal,which works as the substance basis of relieving IBS.All the works here are the foundation to develop new effective drugs for IBS.
