| Objective:To observe the effect of Modified Buzhong Yiqi Decoction for preventing Pneumocystis carinii pneumonia (PCP) and the influence on TNF -αand IL-2 expression about rats, preliminary explore the immune regulation and mechanism on PCP rats and provide experimental data or theoretical basis for the Chinese medicine to fight AIDS and other related diseases.Methods:70 female wistar rats was selected,weighing 180 to 220g.The rats were randomly divided into five groups:normal control group (A), the control group of the PCP model (B),Chinese herb- prevented group(C), Chinese herb-treated group(D),TMP-treated group (E).Except Group A,others were PCP model,Model was established according to the documents,that was, hypodermic injecting into thigh inside of each rat with dexamethasone 1mg/time,twice a week, 8 weeks successively, besides, adding 1g/L acheomycin in to the drinking water in order to prevent secondum bacterial infection. Group A since the first week were given NS gavage, 2ml/time only, and even to eight weeks;group B weren't given medications; Group C since the first one week, were given the Chinese decoction gavage, 2ml/time only, and even to eight weeks; Group D since the first six weeks were given the decoction gavage, 2ml/time only, Even to three weeks; Group E from the first six weeks were given co-trimoxazole 250mg/kg/d, 2times/d, and even to three weeks. Observed the weight, survival rate ,Lung tissue pathology, the cyst number per field in lung smear slice , the level of serum tumor necrosis factor (TNF-α) and interleukin-2 (IL-2), PC trophozoites and ultrastructural of cysts in electron microscope.Results:The survival rate, the average weight after experiment and the decrease of lung smear slice cysts of Chinese herb-prevented and Chinese herb - treated groups with significant differences (P<0.05)than that of PCP model group (P<0.05); Lung tissue pathology slice demonstration Chinese medicine in the prevention group and the treatment group rat Lung organization inflammatory response were significantly lighter than PCP model group;serum TNF-αand IL-2 concentration were higher than PCP model group(P<0.05). By the electron microscope, the damaged lung tissue lesions of Chinese medicine in the prevention and treatment group rats were lighter; the pneumocystis carinii had remarkable vacuolated type changes, Trophozoites dissoluted and fractured.Conclusions:1.Modified buzhongyiqitang had a role in the prevention and treatment of rats'PCP.2.enhanced the expression levels of TNF -αand IL-2 in PCP rats'serum. |
