| Objective:observe the influence and pathomarphological changes of YiQiHuoXue compound decoction on CRP,ET-1,PDGFmRNA's gene expression.discuss Yiqihuoxue method's mechanism and effect target in treating restenosis after PTCA.Method:choose 40 New Zealand white rabbits,and randomly divide them into 2 groups after adaptive feed for one week.Four of them were fed with normal food named normal control group.36 were fed with high cholesterol food named operation group.The later group was performed with balloon injury on carotid artery.After 4 weeks,the opertation group was randomly divided into 4 groups which were model group,XueZhiKang group(Chinese medicine control group),simvastatin group(west medicine control group),YiQiHuoXue compound decoction group(therapy group),according to the level of lipid,weight,etc.The operation group was fed with drug separately,the index of each group was tested in week0,week 4,week8. At the end of 8 weeks,test the CRP,ET-1 in the central artery of ear with empty stomach,and take a tissue to test PDGFmRNA's gene expression.observe the pathomarphological changes under light and electron microscope.Results:TC,TG,LDL level were obviously increased,HLD was decreased.ET-1 and CRP have a higher level,the expression of PDGFmRNA is higher in model group.The difference is obviously compared with normal control group. TC and LDL of the therapy group decreased,the level is more or less compared with Chinese medicine control group(P>0.05),which is different from west medicine control group,but it has no obvious differences in increasing HLD and decreasing TG(P>0.05). The therapy group's CRP was decreased obviously ,campared with Chinese medicine control group(P<0.05),but less than west medicine control group(P>0.05) .The therapy group decreased ET-1,it has no difference from Chinese medicine control group(P>0.05),close to normal group. therapy group,Chinese medicine control group,west medicine control group all could decrease the expression of PDGFmRNA, there is no difference between each group,and it has no difference with normal control group.Each group fed with drug gets improvement on pathological structure of carotid artery tissue,close to normal.Conclusion:1. YiQiHuoXue compound decoction has effect of modulating blood lipid,and it can decrease TC,LDL,the later effect has difference with west control group,more or less in increasing HLD and decreasing TG,compared with west control group.It shows the decoction can decrease the injury of abnormal metabolism of lipid on structure and function of vascular endothelial cells.2. YiQiHuoXue compound decoction can decrease CRP,EF-1's content,shows that it can release the inflammation of endothelial cells,inhibit the formation of thrombus and proliferation of smooth muscle,improve the progress of restenosis after PTCA.3. YiQiHuoXue compound decoction can modulate the expression of PDGFmRNA,the effect is more or less with west control medicine.It shows the decoction can inhibit the proliferation of smooth muscle,also will do good to restenosis after PTCA. |
