As society lifestyle changes and population ageing,cardiovascular and cerebrovascular diseases have become one of the biggest threats to human health. Because of the overall concept,determination of treatment based in pathogenesis of TCM,the complexity of the chemistry components of traditional Chinese medicine and compounds and the diversity of mechanisms,the therapy of ischemic stroke has its own character.Tongmai Xingnao dripping pills(CD) consists of scutellarin,Chuanxiong volatile and other constituents,as a innovative Chinese medicine of the treatment of ischemic stroke.However,because of the indistinctness of its effective substances,the quality control has some problems.Objective:The paper,according to the drug absorption,distribution and metabolism, illuminates the effective material foundation of the treatment of ischemic strokeMethods:1 With the method of HPLC gradient elution to analyze the blood component of CD,the principal blood components as an index were studied the pharmacokinetics of CD,using statistical software DAS2.1.1 to fit pharmacokinetic parameters.2 MTF automated colorimetry was used to study the survival rate of cerebral cortex nerve cells of the scutellarin and rhizoma Chuanxiong volatile oil in vitro.3 With the method of co-cultivating brain microvascular endothelial cells and astrocytes,the chemical constituents of the blood-brain barrier were studied.Results:1 The results showed that after rats intragastric administration of CD,the main blood components was scutellarin,concentration-time curve became two cabinet open model,and T1/2 was 2.925±0.706 h,Tmax 4.400±0.548 h,Cmax 0.638±0.260 mg·L-1, AUC0-18h 4.528±2.18 mg·L-1·h,CL/F 3.140±1.910 L·h-1·kg-1,V1/F 1.404±1.306 L·kg -1 and MRT0-18h 7.187±1.055 h. 2 As compared to blank group,scutellarin promoted the survival rate of cells in vitro only in high concentrations(P<0.01),and in lower concentrations,rhizoma Chuanxiong volatile oil significantly promoted cell survival rate(P<0.01).3 Ligustilide could permeate through blood-brain barrier model in vitro,and scutellarin could not.And ligustilide in the compounds had increased nearly 40%, compared to rhizoma Chuanxiong volatile oil in solo applianceConclusion:In this paper,the analysis of the blood constituents and the blood-brain barrier of CD has revealed the nature of pharmacokinetics and defined that the scutellarin and rhizoma Chuanxiong volatile oil could pass through the blood-brain barrier to protect cerebral nerve and promote the open of the barrier to raise the crossing rate of ligustilide,without scutellarin permeated,which might protect nerves from other ways. the effective components of CD have been initially clarified.The improvement of the scientific quality standards has laid the foundation of interpretation.Innovation:In this paper,co-cultured brain microvascular endothelial cells and astrocytes in vitro has estimated whether the active ingredients of CD could pass through the blood-brain barrier.Combined with the modern techniques analysis of GC-MS and others to analyze the ingredients and identify the structure,a new method of the effective and rapid screening of the chemical composition of traditional Chinese medicine in the blood-brain barrier permeability has been applied,and the effective compound of Chinese medicine and its treatment of ischemic stroke have also been revealed to therapy ischemic stroke.
