Objective: The aim of the present study was (1) To investigate the effects ofghrelin in hippocampal CA1 area on gastric motility and its mechanism by which hypothalamic ghrelin is involved in the regulation of gastric motility; (2) To study the influence of ghrelin on the calcium-activated K~+ current of guinea pig gastric myocytes, and the function of motilin with the regulation of NO; (3) To explore the effects and mechanisms of Ghrelin on the learning and memory in rats.Methods: (1) measure the gastric motility in freely moving rats to exam theeffects and the mechanism of microinjection ghrelin into hippocampal CA 1 area on gastric movement.(2)Patch clamp experiment: Acute dissociation of guinea pig gastric myocytes and observe the influence and mechanism of ghrelin on calcium-activated K~+ current of gastric myocytes using whole-cell patch-clamp techniques.(3) Step-down test and morris water maze were used to test the effects of learning and memory ability by means of microinjecting ghrelin into hippocampal CA1 area in rats.Results: (1) The amplitude of gastric motility was decreased by miroinjection ofthe Ghrelin into hippocampus CA 1 area (P<0.01). The effect was increased by L-NAME (P<0.05) and was decreased by L-AA (P<0.05). (2)The results of path-clamp: Ghrelin can inhibit the calcium-activated K~+ current of guinea pig gastric myocytes (P<0.05). (3) Ghrelin can improve the learning and memory ability of rats (P<0.05).Conclusion: Ghrelin can decrease the amplitude of gastric motility, inhibit thecalcium-activated K~+ current of guinea pig gastric myocytes and improve the learning and memory ability of rats. And all of the functions are implemented by No pathway.
