Topiramate In The Pentylenetetrazol Induced Epileptic Rats Body Weight And Serum Gastrin And Hypothalamus Of Orexin Influence

Objective :To study the effects of topiramate on Serum level of gastrin,the expression of orexin A in hypothalamus,and their relationship with weight changes in pentetruzole-induced epileptic rats;To explore the mechanism in which topiramate causes the descent of the rats'weights and .provide the basic theory.Methods:according Diehl's ways to make chronic epilepsy animal modle.120 wistar (28days) rats were divided into 4 groups randomly:blank group(n=30). TPM compared group(n=30),PTZ model group(n=30),and TPM treated group(n=30).Blank group :intraperitoneal injection of 0.9%Nacl 3.5ml/kg;TPM compared group:input of TPM 40mg/kg through ventriculus duct;PTZ model group : intraperitoneal injection of pentetruzole 35mg/kg;TPM treated group:input of TPM 40mg/kg through ventriculus duct 30 minutes before intraperitoneal injection of pentetruzole 35mg/kg.Once a day from 8:00-10:00 am. recording the seizure degree.If continuousely the sum of degree 4 and degree 5 add up to 3,it will be called successful.Weight changes were measured every 3 days.The level of serum gastrin and the expressing of orexinA in hypothalamus were surveyed,using the method of radioimmunity and immunohistochemicaliy method respectively on the 2nd,the 4th,the 6th weekends.Results:TPM compared group and TPM treated group showed the descend of the rats'weights,comparing with the other two groups on the 3rd weekend and fell to the nadir on 4th weekend (P<0.05); on 4th weekend:the level of serum gastrin descended clearly,comparing the TPM compared group with the blank group and the TPM treated group with the PTZ model group;also the PTZ model group with the blank group and the TPM treated group with the TPM compared group (P<0.05);and orexinA in hypothalamus descended clearly,comparing the TPM compared group and the TPM treated group with the PTZ model group and the blank group(P<0.05).on the 6th weekend:the level of serum gastrin descended clearly,comparing the TPM treated group with the blank group and the TPM compared group,and the PTZ model group with the blank group (P<0.05);the level of serum gastrin did not show significant change, comparing the TPM compared group with the blank group and the TPM treated group with the PTZ model group(P>0.05).orexin-A in hypothalamusdid not show significant change ,comparing the TPM compared group,the TPM treated group with the PTZ model group,the blank group;also the PTZ model group with the blank group and TPM treated group with the TPM compared group(P>0.05).Conclusion:topiramate can lighten the rat'weights have something with descending the expression of orexinA in hypothalamus,but have no directly anything with the descending the level of serum gastrin.
