| Objective: Major feature of nephrotic syndrome is a large number of proteinuria , The renal glomerulus mesoderm cell (i.e. podocyte ) is damage in pathology, more and more studies confirm that the damage cells promote glomerulosclerosis . This study is to explore the process of the rat glomerulosclerosis, the abnormal change of endogenous angiogenesisand and pathological changes .Methods: Healthy Wister rats 64, the average body weight of Healthy Wister is 150-200 g, from the University of Jiamusi Animal Experimental Center and divided into two groups in random(1) Nephropathy group 32. (2) The control group 32. Modeling methods of nephropathy in rat are as follows: rats are anesthetized with Chloral Hydrate, the right kidneys of rats are removed . Adriamycin(ADR)-induced nephrotic rat models were established single injection of 7.5mg/kg adriamycin via the tail vein. (ADR dilution of 2 mg / ml normal saline) after 7 d .All rats were kept in standard cages to be fed and water with standard feed . The control group is fed with normal feeding. 8 ADR rats and 8 control rats were sacrificed at 1,2,4and7weeks after adriamycin injection respectively. 24hr urinary protein, serum cholesterol, serum triglyceride and renal function of rats were measured by routine biochemical method. They are in cage metabolism three day before killed to adapt to environment. Injecting normal saline into abdominal aortic and removing kidney . The expression of Ang-1,Ang-2 mRNA in renal at the same time points were detected by reverse transcriptase polymerase chain reaction (RT-PCR).Results: (1) In ADR rats, urinary protein and the level of triglyceride and in serum increased significantly 14th day, at same time, albumin in serum significantly significantly(P<0.01).Changes of these indices reached the peak level at 49th day. No significant changes are in BUN, Cr. (2) Ang-1 mRNA in ADR was significantly downregulated at2,4 and 7weeks, compared with control group at other time points (P<0.05). Ang-2 mRNA in ADR renal was not detected at 1week. Ang-2 mRNA in ADR was significantly upregulated at2,4 and 7weeks, compared with control group at other time points (P<0.05). (3) Ang-1mRNA and Ang-2mRNA are different in ADR at different time point(P <0.05).Conclusion: 1,Abnormal changes appear in adriamycin nephropathy of rats during glomerulosclerosis.Ang-1mRNA was downregulated and Ang-2mRNA was upregulated. The changes which may be the compensative mechanism under diseases conditions, exists in adriamycin nephropathy . 2,In adriamycin nephropathy kidney, the change of endogenous angiopoieties is partly connected with the renal capillary damage ,Ang-1 and Ang-2 imbalance involved in glomerularsclerosis.3,Angiopoietin upregulation of gloerular part increased cateterization of Urine protein,these changes are related to podocyte damage. |
