Objective:To detect the expression of erythropoietin and erythropoietin receptor(EPOR) in the patients with myelodysplastic syndrome(MDS) and its clinical implication. At the same time,we also explore the relationship of serum erythropoietin and anemia in patients to supply a theoretical basis for the cytokines therapy in anemia patients with MDS.Methods:Samples were collected from patients with myelodysplastic syndrome from May to October in 2007,diagnosed according to the WHO classification. Thirty patients and twenty controls were investigated: 18 men and 12 women,aging from 39 to 81,the average is 56 years old,including 16 MDS-RCMD,8 MDS-RA,3 MDS-RAS and 3 MDS-RAEB in MDS group. The level of EPOR mRNA and serum erythropoietin were detected in 30 patients using reverse transcription polymerase chain reaction and sandwich ELISA respectively.Results:EPOR was expressed in all 30 patients and 20 controls. The average quantity of EPOR mRNA expression in MDS was 0.42±0.10,significantly lower than that of control group(0.48±0.11)(P<0.05). The serum level of EPO were 25.68±22.49 mIU/ml in MDS and 10.57±9.89mIU/ml in controls.It was higher in patients than controls(P<0.01),and there was negative correlation between the level of sEPO and Hb(r=–0.737,P<0.01).Conclusions:(1) EPOR was expressed in the patients with myelodysplastic syndrome,the expressive level was lower. (2) The serum levels of EPO in MDS patients were higher than that in controls. (3) Recombinant human erythropoietin is important in treatment of anemia caused by MDS,it can alleviate the anemia caused by MDS.
