Ischemic stroke is caused by blood and oxygen of deficiency due to brain blood supply disorder, which cause local brain tissue ischemic necrosis and cerebromalacia. It is a serious harm to human health and quality of life, the rates of attack, mutilation and death are high, induces heavy burden. Large researches show that acupuncture for ischemic stroke can reach good therapeutic efficacy, but researches about acupuncture for precaution and healing of ischemic stroke, and acupuncture on the influence of EPCs are few. The steps of EPCs from marrow to blood are divided into motivation, migration, homing and differentiation, then join neovascularization in the local region. VEGF have motivation function to EPCs from marrow, it works on the surface of EPCs, induce the growth, then runs into blood. This research uses the model of (MCAO/R), observes the influence of pretreatment of electro-acupuncture to EPCs and VEGF, studies the mechanism of treatment of acupuncture.Objective: To observe the influence of electroacupuncture pretreatment to the EPCs and VEGF in the marrow and peripheral blood, to investigate the mechanism of electroacupuncture pretreatment in relieving brain ischemic reperfusion injury.Methods: Divided the rats into treatment group, model group, sham operation group and normal group by sortition randomization method. After the models were made, used the flow cytometer to detect the EPCs in the marrow and peripheral blood, and detect the VEGF in the marrow and serum by ELESA method, in 12h, 24h and 48h. Statistic the results by SAS software.Results:(1)in the time of 24h and 48h, EPCs in marrow of treatment group is higher than model group, the more percent of EPCs the longer time;(2)in the time of 12h,24h and 48h, EPCs in serum of treatment group is higher than model group, the more percent of EPCs the longer time;(3)in the time of 24h and 48h, VEGF in marrow of treatment group is higher than model group, the more percent of VEGF the longer time,(4)in the time of 12h, 24h and 48h, VEGF in peripheral blood of treatment group is higher than model group, the more percent of VEGF the longer time.Conclusion:(1)MCAO rats'content of EPCs and VEGF in bone marrow and peripheral blood is higher than narmal rats';(2)electroacupuncture pretreatmented MCAO rats'content of EPCs and VEGF in bone marrow and peripheral blood is higher than unpretreatmented MCAO rats';(3)electroacupuncture pretreatment can increase the content of EPCs in bone marrow and peripheral blood, so electroacupuncture pretreatment can promote the motivation and migration of EPCs;(4)electroacupuncture pretreatment can increase the content of VEGF in bone marrow and peripheral blood, so electroacupuncture pretreatment can promote the motivation and migration of EPCs through VEGF increasing;(5)electroacupuncture pretreatmented MCAO rats'EPCs and VEGF content are the more the longer time; (6) pretreatmented acupuncture has the functions of dredging meridine, activating blood, relieving stasis and growing fresh blood, opening orifices.
