Amoxicillin,omeprazole and tinidazole combined to use is a common project which cures digest ulcer in clinic.Ampicillin sulbactam and ornidazole combined to use is a common project to cure anaerobic infection in clinic.The present study investigated the compatible stability of amoxicillin sodium,omeprazole sodium,tinidazole,ampicillin sodium and sulbactam sodium and ornidazole with 8 infusion solutions.Besides,the compatible stability of amoxicillin sodium, omeprazole sodium and tinidazole in SLR,SLS,the compatible stability of ampicillin sodium and sulbactam sodium with ornidazole were investigated.The aim of the present study was to provide a reference for clinic.The present study provided methods to determine the contents of amoxicillin sodium, omeprazole sodium,tinidazole,ampicillin sodium sulbactam sodium and ornidazole by high performance liquid chromatography.To establish a method for the determination of amoxicillin sodium,omeprazole sodium and tinidazole by RP-HPLC,the separation was performed on a C18 analytical column(4.6 mm×150 mm,5μm,ZhongHuiDa)with the mobilephase consisting of acetonitrile-pH7.6 phosphate buffer(30:70)with time gradient elution mode.The detection was set at 285 nm.To establish a method for the determination of ampicillin sodium,sulbactam sodium and ornidazole by RP-HPLC,the separation was performed on a ZORBA*SB-C18 analytical column(4.6 mm×250 mm,5μm,Agilent)with the mobile phase consisting of acetonitrile-0.02 mol·L-1monopotassium phosphate(15:85)with time gradient elution mode.The detection was set at 230 nm.The pH,quantity of insoluble particulate and appearance of mixtures were also observed to investigate the compatible stability of the five medicines.The results were as follows:Amoxicillin sodium and R2A,R4A,M3A were compatible in 1 hour.It was compatible with other 5 infusion solutions in 6 hours;Omeprazole sodium and SLS,SLR were compatible in 6 hours,whereas the other six infusion solutions;Ampicillin sulbactam and M3A,M3B,R4A were compatible in 1 hour.It was compatible with other 5 infusion solutions in 6 hours;Tinidazole was compatible with 8 infusion solutions in 6 hours; Ornidazole was compatible with 8 infusion solutions in 6 hours;Amoxicillin,omeprazole and tinidazole were compatible in SLR,SLS in 6 hours;Ampicillin sulbactam and Ornidazole were compatible in 6 hours.The present study investigated the catabolite of omeprazole because omeprazole sodium is degtaded in acidic infusion solution.One catabolite was separated by semi-preparative high performance liquid chromatography.The ultraviolet absorption spectrogram provided two absorption peaks:214 nm and 300 nm.Mass spectrometric detection was carried out by Thermo Finnigan LCQ Advantagemass spectrograph.ESI(Positive mode)was adoped.Analysis showed the molecular weight of this compound was 329.This compound was speculated as 5-Methoxy-2-(((4-methoxy-3,5-dimethyl-2-pyridinyl)methyl)thio)-1H-benzoimidazole, preliminarily.
