Background and Purpose:For a long time, the repair of peripheral nerve injury for orthopedic medical workers are a problem. Clinically, for the treatment of peripheral nerve injury in particular long injury or defect in the way, autologous neural transplantation is still the most reliable means of treatment, but for the loss of nerve function and nerve repair for a limited length and diameter is insurmountable shortcomings. Silicone tube, and other synthetic materials and autologous skeletal muscle, vein-derived biological material to guide the nerve regeneration, but its structure and the basement membrane of different axon regeneration poor clinical application of little value, nerve allograft transplantation can be overcome these shortcomings, but there is rejection problems. The purpose of this study: Pre-infusion through the tail vein of the ultraviolet radiation of donor spleen cells and the sciatic nerve allograft transplantation, determination of peripheral blood T-lymphocyte subsets, repair of peripheral nerve defects feasibility. For the repair of peripheral nerve defects provide a new approach.Methods:UVB irradiation donor spleen cells Wister rats via the tail vein infusion to the receptor SD rats, experimental divided into three groups: Group A for autologous nerve group, Group B for peripheral nerve transplant team, in group C tail Pre-venous infusion by UV irradiation of donor spleen cells of allogeneic transplantation. Seven days for the rat sciatic nerve transplantation. Postoperative neurological lectrophysiology, histology and peripheral blood T-lymphocyte subsets detection.Results:After seven days immunological tests showed that peripheral blood T-lymphocyte subsets inspections, in group B than in group C of CD8 + T cells to increase the percentage of CD4 + T cells to reduce the percentage of CD4/CD8 ratio of small groups The difference between statistical significance (P <0.01). Group A and C, the difference was significant (P <0.05). After eight weeks, the motor nerve conduction velocity, histological examination, in group B than in group C, the difference was significant (P <0.01) in group A and group C had a significant difference (P <0.05).Conclusion:To the tail vein injection of the antigen gene differences spleen cells can be induced peripheral nerve transplant on the specific immune tolerance.
