ObjectiveThe substance of Meridian is the key issue on which has been focused domestic and foreign scholars,but to date it has not yet been clearly explained.From the description of Ancient Chinese medicine book,we can know that the meridian is the channel running the channels qi and blood,"Acupuncture Masterpiece" explicitly pointed out:"The meridian is the road running qi and blood ","The qi is the general for blood,and the blood is the mother for qi".Therefore the knowing about qi and blood can help us to understand the essence of the meridian.The channel qi is resulted from the water-absorption by spleen and stomach and the pure atmosphere respired by lung,which through the meridians transports to the whole body and to provide nutrition for the viscera,and to participate in conditioning the function and activities of zangfu.The function of regulating zangfu and the the treatment for diseases achieved by acupuncturing.Acupuncture can treat diseases is due to the function of "adjusting qi".Such as "Lingshu·cijiezhenxie"said that the key of acupuncturing is regulating the qi."Acupuncture Masterpiece" said:"would rather miss the time,than miss the qi." "Suwen·baomingquanxinglun" said that the channel qi has been arrived, which should be maintained cautiously".The descriptions above all are illustrating the sinificance of qi in the treatment for diseases.So,qi and blood in the human life is a very important material.Modern physiology has confirmed that the oxygen is identical to the qi and blood in TCM plays an important role in the course of human life,which participates in various physiological responses.The energy suppled for human body system is resulted from the biochemical reactions by oxygen and three major nutrients of sugar, fat,protein,which are intaken by food.Oxygen metabolism in human life activities play a vital role.According to the understanding of oxygen metabolism in modern physiology and the Chinese meridian theory,Pro.Liang posed the hypothesis about the substance of channel qi and meridian in1990s[1].Our experiment guided by the hypothesis,through acupuncturing the acupoints Chize(LU5)on Lung Meridian of Hand-Taiyin and Quchi(LI11)on Large Intestine Meridian of Hand-Yangming to detect the blood gas analysis index(PO2,CaO2,SaO2%,PCO2,TCO2,HCO3-,PH)and the oxygen metabolism indicators(SOD,GSH-Px,MDA)contents.By utilizing the experimental method to disscuss the relativity between the modern physiology and the qi in TCM and channel qi,in order to gradually reveal the substance of channel qi and meridian and lay the foundation for real meridian.MethodsWe select 20 normal young men and women as the subjects for the study,and the male to female ratio is 50:50.The ages of the subjects are from 23 to 29,the average age is 25;they are divided into two groups(Lung Meridian of Hand-Taiyin groups and Large Intesine Meridian of Hand-Yangming groups),and each group includes 10 cases.We utilize the method of PingBuPingXie,compared with the blood gas analysis index(PO2,CaO2,SaO2%,PCO2,TCO2,HCO3-,PH)and oxygen metabolism(SOD,GSH-Px,MDA)on acupoints and non-acupoints before and after acupuncture.The experiments are carried out in Chinese medicine hospitals in Hubei Province and chemical and biological testing laboratory in Hubei college of TCMThe first experiment:the affect to the analysis of blood gas content of indicators on human acupoints and non-acupoints.1.Specimens collected:the blood collection sites on subjects in Lung Meridian of Hand-Taiyin groups before acupuncture are Shaoshang(LU11)and the control points (the end of thumb ulnar,from the angle 0.1-inch nail);the blood collection sites on subjects in Large Intesine Meridian of Hand-Yangming groups before acupuncture are Shangyang(LI1)and the control points(end ulnar index finger,nail angle from the 0.1-inch).Regular disinfection,blood collection needle acupuncture bleeding immediately after the draw with micro pipette into centrifuge tube in the anticoagulant heparin.2.Detection of indicators:in the whirlpool on a blending and mixing,standing five minutes,using micro-glass capillary draw 40 ul of whole blood,blood gas analysis sent to AVL measured PO2,CaO2,SaO2%,PCO2,TCO2,HCO3-,PH.The remaining blood samples placed in a centrifuge 3000 / min from heart after 15 minutes standing for one minute.From the liquid supernatant in the reagent in the refrigerator at -30℃preservation,be prepared for the next experiment.3.Acupuncture:the subjects in Lung Meridian of Hand-Taiyin groups are acupunctured Chize(LU5),and the subjects in Large Intesine Meridian of Hand-Yangming groups are acupunctured Quchi(LI11)for 30min,all of them acupunctured by TiChaBuXie method.4.The methods of specimen collection,index detection are identical to step 1 and stept 2.The second experiment:The affect to the SOD,GSH-Px,MDA contents on human acupoints and non-acupoints.1.Take out the specimen stored in the refrigerator in the first experiment,and in accordance with the kit manual steps strictly,we are using xanthine oxidase method to detect SOD,colorimetric method to detect GSH-Px,and thiobarbituric acid method to detect MDA.2.721 spectrophotometer to detect the absorption of the three indicators,and based on targeted reagent concentration formula to calculate the activity of SOD, GSH-Px,and the concentration of MDA.Statistics manipulate:measurement data are expressed in the form of mean±standard deviation((?)±S),the date analyzed by SPSS15.0 statistical data processing software, the comparison between the two groups with matching t-test. Results1.The impact on blood gas analysis indicators by acupuncturing.1.1 Before acupuncture,there are no significant differences of the contents of PO2, CaO2,SaO2%,PCO2,TCO2,HCO3-,PH on acupoints compared with the non-acupoints(P>0.05).1.2 After acupuncture,the contents of PO2,CaO2 on points are lower than non-points;while the contents of PCO2,TCO2,HCO3-,PH on acupoints are higher than non-acupoints.The difference is significant(P<0.05).The SaO2%have no statistical difference(P>0.05).1.3.The contents of PO2,CaO2 after acupuncture are lower than before acupuncture on points;the contents of PCO2,TCO2,HCO3-,PH after acupuncture are higher than before acupuncture,the differences are significant(P<0.05).The SaO2%have no statistical difference(P>0.05).1.4.There are no significant differences of the contents of PO2,CaO2,SaO2%,PCO2, TCO2,HCO3-,PH before and after acupuncture on non-points(P>0.05).2.The impact on oxygen metabolism by acupuncturing.2.1 Before acupuncture,compared with non-acupoints,the contents of SOD,GSH-Px, MDA in no significant difference(P>0.05).2.2 After acupuncture,compared with non-acupoints,the contents of SOD,GSH-Px are higher than non-acupoints,the differences have statistical significance(P<0.05); the contents of MDA are lower than non-acupoints,the differences have statistical significance(P<0.05).2.3 Before and after acupuncture,the contents of SOD,GSH-Px on acupoints are conspicuously higher than before acupuncture,and the contents of MDA declined sharply after acupuncture,the differences have statistical significance(P<0.05).2.4.Compared with the non-acupoints,the changes of the contents of SOD,MDA on non-acupoints are significantly(P<0.05),but the changes of the contents of GSH-Px have no significance(P>0.05). Conclusions1.The impact on oxygen metabolism relative indicators on remote acupoints and the non-acupoints by acupuncturing are significantly different,it is suggested that the channel qi maybe closely associated with the oxygen metabolism.2.The mechanism of regulating channel qi realized by acupuncture maybe through adjusting the state of oxygen and its metabolites3.SOD,GSH-Px may be one of the substance of meridian and channel qi.
