Objective:Bradycardia is one of the common cardiovascular disease among the elder people,the treatment is lack of specificity in clinical.The patients who have no symptoms don't need to take medicine,while the serious have a temporary curative effect but recur easily.The long-term use of westem medicine had a great deal of side-effect, currently it has been proved that antiarrhythic drugs mostly had the side-effect of causing arhythmia,moreover,the serious bradycardia can cause Aase's syndrome, even sudden death,so these patients have to use cardiac pacer.The cardiac pacer costs a lot for the serious patients and brings them tremendous burden both in economy and life.Acupuncture has the unique advantage in cardiovascular disease,and can decrease the toxic and side-effect of the drug to a great exent.We made the Bradycardic Model of rats by intragastrically gevenβ-R blocker(Propranolol),to observe the effect of the electro-acupuncture(EA)at neiguan acupoint on the heart rate,ECG and the content of The SOD and MDA in rats with bradycardia.Moreover,we evaluate the therapeutic efficacy,and approach its mechanism of action,try to make a scientific basis for electro-acupuncture treatment of bradycardia.Methods:40 SD rats were selected.Each rat's body weight was from 250 to 300g.They were randomly divided into four groups,each group with 10rats:normal group,model group,EA on neiguan treatment group(EA treatment group)and EA on lieque control group(EA control group).Each group of rats were embedded electrode underneath the four limbs,connecting and recording with bio-computer.Normal group rats were not provided treatment.The other three groups were made Bradycardic model which were intragastrically geven 0.2%Propranolol Hydrochloride suspension.EA group were treated by EA with condensed and rarefactive wave of 2Hz/15Hz after the model was sucessfully made.The treatment was continued for 20 minintes.After that,we collected the specimens,and detected the content of SOD and MDA in myocardium with xanthine oxidase method and thiobarbituric acid method separately,and analyzed Statistically.Results:1.Gavage of the model,the rats' heart rates decreased obviously than normal group rats'(p<0.01),but they were enhanced after treated by EA on neiguan acupoint, and closely approached to the normal group,compared with model group,there was very significant difference(p<0.01).The heart rate increased after treated by EA on lieque acupoint,but its effect was worse than EA treatment group,compared between the two groups,there were significant difference(p<0.05).2.The ECG showed that the Q-T interval of model group was prolonged obviouly (P<0.01),which was abbreviated obviously after treated by EA on neiguan acupoint (p<0.01),approximately approached to the normal group;Compared with model group,the Q-T interval of EA control group was also abbreviated,there was significant difference(p<0.05);the Q-T interval of EA treatment group was obviously shorter than EA control group,and there were significant difference between the two groups (p<0.05).3.Compared with normal group,the content of SOD in model group rats' myocardium decreased obviousely,while the content of MDA increased greatly(p<0.05), this implied that both the content of SOD and MDA in rats' myocardium had abnorma lity variation in Bradycardia.Compared with model group,the content of SOD in EA group rats' myocardium increased obviousely,while the content of MDA decreased greatly,but EA treatment group had of more significant changes(p<0.05).Conclusion:1.The therapy of EA on neiguan acupoint obviously improved the rats' heart rate, and its effect was superior to that of EA on lieque acupoint,indicating that EA can antagonize Bradycardia.2.The contents of SOD decreased greatly and the contents of MDA increased obviousely in myocardium in Model group,which indicated that the content of SOD and MDA had abnormality variation in Bradycardia.The contents of SOD increased greatly and the contents of MDA decreased obviousely in myocardium in EA treament group,which indicated that EA therapy has obviouse regulating effect for the contents of SOD and MDA in rats' myocardium. |
