The purpose of this research is to explore the best technical conditions and microemulsion preparing technology in extraction and refine of Angelica,the research of microemulsion of Angelica in pharmacodynamics,and the relative bio-availability of microemulsion of Angelica a kind of traditional Chinese medicine in different administration routes in Tongluojiunao micro emulsion preparation, a traditional Chinese medical formulae., In this experiment we found that:1. Research on refine technology on recirculation of angelicaOptimization of recirculation technology of Angelica: taking times of water added(A), time of abstraction(B), frequency of abstraction(C) and alcohol extracting as the factors, three levels for each, adopting the Li (34) Cross experiment,moreover, yield of ferulaic acid as the indicators we screen the best technical conditions for recirculation technology of Angelica. The conclusion in this cross experiment is: taking the content of ferulaic acid as the indicator, the sequence of each level is one> two> three,the analysis of variance shows that: all the three level, one, two and three have the significance. Therefore, the best technology of recirculation of Angelica is: taking times of water added is ten, time of abstraction is an hour and half, frequency of abstraction is three times and sedimentation of alcohol is 70%.2. Research on preparation technology of micro emulsion of angelicaResearch on empty emulsion prescription: taking tween-80 as the surface acting agent, alcohol as the assistant surface acting agent, angelica oil as the oil phase. Mixing tween-80 and alcohol by the volume ratio (Km) of 4:1, 3:1, 2:1, 1:1 andl:2, and then mixing the mixed liquor with angelica oil by quality ratio respectively, stirred to well distributed, stirred on the magnetic stirring apparatus and titrated each mixture by deionized water while stirring to prepare the micro emulsion, the system changed from turbid to transparence shows microemulsion formed. Drawing the pseudo-three element phase diagram. The micro emulsion ratio is confirmed.But we allow for many factors in animal experiment,for example, dosis tolerate of animal and dosage, concentrated solution of water solution of Angelica take the place of deionized water. Observed under the transmission electron microscope, inner phase is round and well distributed.3. The research of pharmacodynamics of micro emulsion of angelicaThe influence of angelica microemulsion to rat clotting time and whole blood viscocity. The impact of angelica microemulsion large,medium and small three angelica emulsion to the rat clotting time and whole blood viscocity. Angelica microemulsion large,medium and small three-dose group has the remarkable difference. As the dose increasing and the effect of anticoagulated blood strengthens, displays the good does-effect relationship. Compared with the decoction of angelica, microemulsion can prolong the clotting time significantly. And the performance for the three-dose group compared with liquid water to cook, there is extremely remarkable difference. The blood viscosity of reduce stagnation of blood model rat stagnation of blood, compares with the model group, there is the remarkable difference. In addition, under the low shear rate, the difference between high-dose group microemulsion lower blood viscosity effect and the liquid water to cook has the remarkable difference, under the high shear rate, high-dose group microemulsion lower blood viscosity effect is stronger than the flow of water to cook and extract group, statistics comparison has the remarkable differrnce.4. Research on relative bioavailability of angelica in different administration routeDetermination of Angelica blood concentration in rabbits in different administration route: taking ferulaic acid as the indicator, peak amplitude of Carbinoxamine density of water p.o. is after thirty minute of administration, and attenuates by degrees . Velocity is very quick.; experiment shows: the bioavailability of micro emulsion is 190.56%.
