As a serious threat to people's lives and health,annually, the incidence number of cancer is around 1.6 million in China, which is greater than the number of cardio-cerebral vascular disease. And 90% of the patients died of tumor metastasis, the first cause of death. Metastasis is the characteristic performance of tumor, and it is also a malignant behavior which is the main reason for unsuccessful treatment. Therefore prognosis of cancer patients depends on whether the transfer can be successfully controlled or not. Tumor metastasis includes hematogenous metastasis,lymphatic metastasis and violation of cultivation, thereinto, hematogenous metastasis is the main way, and in this process, the interaction between tumor cells and vascular endothelial cells is the most complex and important tache. In recent years, the researches on the mechanisms of anti- metastasis by Chinese traditional medicine have made huge achievements, and it was reported that Berberine can inhibit tumor metastasis. Although we have achieved certain results about the mechanisms of how it inhibits metastasis, there is few reports about direct and dynamic observation of the effect on cells of Berberine. In this study, we will use optical imaging methods to study the effect of berberine(Ber)on invasion and migration of PG cells line from highly metastatic human giant cell lung carcinoma and to explore its mechanism.1.PurposeTo study the effect of Berberine on invasion and migration of PG cells line from highly metastatic human giant cell lung carcinoma by optical imaging methods and to illustrate its mechanism on cellular and sub-cellular level.2.MethodIt is divided into two groups:PG cell group and PG&HUVEC adhesion group, while each group includes control set and Berberine set. PG cells were marked with DPH, then to observe the effect of Berberine on the changes of the fluidity of the membrane of PG cells by Fluorescence Redistribution After Photobleaching(FRAP).Confocal laser scanning microscopy was used to observe the effect on the expression of CD44s and CD54 on PG cells after PG cells being marked with FITC-CD44sAb and PE-CD54Ab.3.ResultsBerberine was shown to inhibit the fluidity of the membrane of PG cells in both PG cell group and PG&HUVEC adhesion group. Berberine reduced the expression of CD44s and CD54 on PG cells in both PG cell group and PG&HUVEC adhesion group.4.ConclusionBerberine has an inhibitory effect on the migration of PG cells,and the effect may be carried out by inhibiting the fluidity of the membrane of PG cells and reducing the expression of CD44s and CD54 on PG cells.
