It's defined that caulis pipers futokadsurae is vine of pepper family's plant martin fig root and stem---Piper Kadsura (Choisy) Owchi in Chinese Pharmacopeia (2005 edition). It possesses the effect of eliminating beriberoid disease and meridian deoppilation, and it can treat anemofrigid-damp arthralgia, arthral soreness, tendon spasm of tendon and vessel, injury from fall. Investigations discover that the distribution range of the plant martin fig root and stem(Piper Kadsura (Choisy) Owchi) is narrow and the plant distributes only in Fujian and Hainan etc. sporadically, so martin fig root and stem is utmostly short of resource. But Piper hancei which is the same family(pepper family) and group(piper) with martin fig root and stem distributes widely in Fujian, Zhejiang, Jiangxi, Guangdong, Yunnan, Guizhou and so on, and have a great quantity in resource. Identity result of caulis pipers futokadsurae on market indicates that the initial plant of caulis pipers futokadsurae which is applied the most widely and has maximal flow flux in most territories throughout China is vine of Piper hancei.We carried out pharmacognostical study on caulis pipers futokadsurae and Piper hancei by all kinds of means. First, we observed quale of caulis pipers futokadsurae and Piper hancei by normal means, and found that their quale are very similar. We observed cross-section and powder of two medical materials under microscope by normal means, and found that character of their cross-section has not much difference and character of their powder are similar. We carried out chemical qualitative analysis on caulis pipers futokadsurae and Piper hancei, respectively take tart alcohol extract of the two and carried out alkaloid precipitation through adding silicotungstic acid, Potassium Heptaiodobismuthate, potassium mercuric iodide to the extract. We discovered that colature of both medicinal substances presented the same masculine phenomenon after adding reactant. It demonstrated that both contain homoplastic alkaloid component. We carried out micro-chemical identification through adding alcohol and dilute sulphuric acid to powder of caulis pipers futokadsurae and Piper hancei, and found that needle crystals appeared on both mountings and solved after adding sulfuric acid. lt proved that structure of both contain calcium oxalate crystals. We put alcohol leaching liquor of both in UV-visible radiation spectrophotometer, and determined ultraviolet spectra in the range of 200--400nm, and found that there are absorption peak near 270nm in ultraviolet spectra of both. It illustrated that key component of both have certain similarity. We took coarse alcohol extract of two medical materials and respectively extracted with ligarine, dichlormethane, acetic ether, n-butanol saturated by water, and diluted every extract liquid 15 times. Then we put every dilution in UV-visible radiation spectrophotometer and determined ultraviolet spectra in the range of 210--700nm. Last we found that ultraviolet spectra of their four location have certain similarity. It certified that component of both have higher similarity. Before carrying out TLC, we extracted component of Piper hancei, caulis pipers futokadsurae and its reference medical material with methyl alcohol and hypersound, filtered, evaporated colature to dryness, added absolute alcohol to dissolve marc in order to obtain test solution. We imbibed 20μl and 10μl test solution, 5μl and 3μl test solution of reference medical material, respectively placed on the same CMC-Na silica gel-G thin layer plate, developed with ligarine(60-90) --acetone(15∶3), open-air dried, and then observed under ultraviolet lamp(365nm), colorated with alcohol of 10% sulfuric acid, observed under ultraviolet lamp(365nm). We found that Piper hancei, caulis pipers futokadsurae and its reference medical material have more principal spots of identical color on corresponding location of TLC. It stated that key component of both have higher similarity.We compared Lignanoid, alkaloid etc. of Piper hancei, caulis Pipers futokadsurae and its reference medical material through HPLC technique. Findings indicated that three pieces of atlas have 15 common characteristic absorptionband. It manifested that Piper hancei has key component of caulis pipers futokadsurae, moreover, result of comparing. peak area indicated the content of the components is higher in Piper hancei than in caulis pipers futokadsurae.We identified caulis pipers futokadsurae and Piper hancei on molecular level through RAPD technique. We extracted general DNA of both with Extraction Kit of Plant genome DNA, and amplified randomly with 20 random primers. At last 13 effective primers were screened to demonstrate the similarity of their amplification band. The resemblance of caulis pipers futokadsurae and Piper hancei can be determined by RAPD which is simply operated efficaciously and quickly.
