ObjectiveThe diabetic rats were induced by Ip of STZ and were fed with high calorie feed irregularly. To discuss the therapeutic effect of TangWeiKang(TWK) on pneumogastric neuropathy of the diabetes rats by observing its influence on motilin (MOT),blood sugar and stomach-intestine driving marker and disclose the mechanism of improving and treating diabetic gastroparesis(DGP).MethodExcept those of the normal control group ,the other rats were used as pathology models of DGP.They were induced by being injected streptozocin(STZ),45mg/kg and their blood sugars were higher than 16.7mmol/L.48 Wister diabetic rats were randomly divided into four groups: model group, TWK high dose treatment group, TWK low dose treatment group,Domperidone treatment group, 12 rats in each group. The control group was feed regularly on standard rats feed.The other four groups were fed on high calorie feed (high sugar and fat), irregularly, eating in odd-number days and fasting in even-numbered days.All the rats drunk water freely. During the experiment period, the appetite, water volume, urine volume, stool character should be observed, and weight the rats, test the urine sugar once every week. Each group was perfused stomach with drugs during establishing the model. The control group and the model group were perfused stomach with saline,1.5ml/100g.TWK high dose treatment group 15ml/kg (48g/kg).TWK low dose treatment group 15ml/kg(12g/kg) and Domperidone treatment group 6ml/kg (0.6mg/kg), once per day, lasted 4 weeks. Recorded the weight and the blood sugar on an un-empty stomach.The day before the last day, Fasting 24h after the last feeding, each group fed carbo-powde suspension by orally taking (activated carbon and acacia each contains 10%),lOml/kg.sampling blood in rats eyeball to measure the motilin content in plasma after 30 minutes.put to death immediately after sampling blood, Calculating the stomach-intestine driving marker, we measured the level of MOT in the blood with radioimmunoassay(RIA).ResultAfter 3 weeks' treatment, the rats of the model group' survival rate were sharply decrease(66.67 %). After 4 weeks' treatment, the rats of the model group compared with the control group, blood sugar Significantly increased(P<0.01),weight obviously decreased,stomach-intestin driving marker sharply decreased(P<0.01), each drug treatment group compared with the model group, the survival rate is respectively as followed: Domperidone treatment group is 75 % ,low and high dose TWK treatment groups are 75 % and 83.33 % .the blood sugar decreased than the model group (P<0.05),weight increased, the high dose TWK treatment group compared with the model group(P<0.05), as the stomach- intestine driving marker and the level of MOT in the blood was concerned, Domperidone treatment group, TWK low and high dose treatment group increased obviously than the model group(P<0.05),there is difference on the stomach-intestine driving marker, the level of MOT in the blood between the high and low dose of TWK treatment group(P<0.05).there is no difference between the TWK high dose treatment group and the control group, on the level of MOT in the blood (P>0.05).ConclusionTWK can ameliorate the symptoms of DGP rats,decrease blood glucose level affectively, improve the peristalsis of gastrointestinal and decrease the level of MOT in the blood,These findings suggested that TWK may have a value in preventing and treating of DGP.
