Objective To investigate the expression of CD147,MMP-2 in ear canal cholesteatoma and middle ear cholesteatoma and their correlation and to determine their roles in the destruction of bone .Material and Method Immunohistochemical method was used to examine the expression of CD147 and MMP-2 in 24 cases of ear canal cholesteatoma ,30 csaes of middle ear cholesteatoma and 20 cases of normal external acoustic meatus skin. Results CD147 was found in basal cell layer,preckel cell layer and stratum granulosum of cholesteatoma epithelium. Positive staining of CD147 was localized in cytolemma and cytoplasm.MMP-2 was found in basal cell layer,preckel cell layer and stratum granulosum of cholesteatoma epithelium and stromal cell.Positive staining of MMP-2 was localized in cytoplasm. There were significant differences about the positive rate of MMP-2 and CD147 between normal external acoustic meatus skin and ear canal cholesteatoma. There were significant differences about the positive rate of MMP-2 and CD147 between normal external acoustic meatus skin and middle ear cholesteatoma.There was a positive correlation between expression of MMP-2 and CD147.There was no significant difference about the positive rate of MMP-2 and CD147 between ear canal cholesteatoma and middle ear cholesteatoma.Conclusions 1.The expression of CD147 and MMP-2 in both ear canal cholesteatoma and middle ear cholesteatoma are significantly higher than that in normal external acoustic meatus skin. MMP-2 and CD147 have a positive correlation in their expression ,which suggest that the two factors maybe act on each other .The higher expression of CD147 might contribute to the increasing secreations of MMP-2 in cholesteatoma. 2. MMP-2 and CD147 are both relate to the destruction of bone in both ear canal cholesteatoma and middle ear cholesteatoma. 3. The effection of CD147 and MMP-2 in ear canal cholesteatoma and middle ear cholesteatoma might be same.
