The First part:Study on preparation and preservation procedure of homologous cardiac valveObject:To investigate the suitable method to prepare and preserve homologous cardiac valve.Methods:Experimental homologous cardiac valve obtain come from donator rabbits,cardiac valve is a series of treated,inclouding crypreserved procedure,crypreserved procedure need a bout 2 weeks.the histology,destroying stress,denaturation temperture,water content,MHC-Ⅱantigen were examined.Results:cardiac valve of donator rabbits were smoothly obtained arotic valve and bicuspid valve keep structural integrality in cut action,cardiac valve biomechanics all around index keep well;A little of endothelial cell have been shed after crypreserved 2weeks.HE(hematoxylin and eosin stain)show a little Inflammatory reaction;morphology display that remnant endothelial cell array according to applanation and monolaye,or Fusiform shape,or polygon, typical arrangement present "slabstone" shape;VG+ET(weigert +van gieson):there are subcutaneouly exposed elastic fibers and collagen fibers,they array regularity,uniformity,order,that consistent with velar suffer force condition;fluorescein isothiocyanate labeling MHC-Ⅱmolecule antigen display specific flavo-green fluorescent light: DAPl(4',6-diamidino-2-phenylindole 2hcl stain):nucleated cell display uniform blue lightConclusion:the method was testified suitable,convenient and available to homologous cardiac valve preparation and preservation,it suggest that that provides a practial useful method for cardiac valve preparation and preservation.The second part:Experimental Study on immunological rejection of homologous cardiac valveObject:To make the model of immunological rejection of homologous cardiac valve:Through analyses of histology,ultrastructure,physics,biochemistry,immunology,biomechanics to study the effect of immunological rejection on cardiac valve;To make use of the inhibitory action of Atorvastatin to MHC-Ⅱfactor about valvular tissue.We aim at reduce the phenomenon of valvular calcify and degeneration thai is caused by immunological rejection,and prolong the lifcspan of biovalve.Methods:To get the cardiac valve after cryopeservation and 16 rabbits were randomized to medicinement group(1mg/kg,3mg/kg,6mg/kg;n:12)and control group(n:4).To embed subcutaneouly the bicuspid valve and aortic valves,then every groups were treated with atorvastatin(1mg/kg,3mg/kg,6mg/kg)and physiological saline.To detect every index after 4 weeks.Results:All animals' wound include medicinement group and control group is healing well.The phenomenon that valves were wraped up is obviously in control group.It is increased significantly(P<0.05)between medicinement group and control group on the histomorphology and immunological rejection of homologous cardiac valve.Other indexs have signment different between every teams(P<0.05).but DAPI after stain have no signment different between every teams.Conclusion:To compare with control group,medicinement group have protection on the histomorphology and immunological rejection.Atorvastatin has the inhibitory action after transplantation of homologous cardiac valve because it decrease the expression of the MHC-Ⅱfacter.
