Objective: The whole layer skin defection rat was used and to evaluate the therapeutic effect of the Chinese herbal compound prescription-Yuhonggao in wound healing. To observe the effect of Yuhonggao on the secretion of substance P (SP) and the reinnervation in wound healing and investigate the mechanism of promoting wound healing.Methods: three full thickness dermal excision wound models with the diameter of 1.8 cm were established on the back of144 healthy male Wistar rat ,which were randomly divided into 3 groups: Yuhonggao group, Jingwanhong group, model group, 48 rats per group. With 6 normal rats as control and Jingwanhong as positive compare medicine. The rats in Yuhonggao group were external applied with Yuhonggao, 0.1 g each wound surface and 2 times a day. The application of Jingwanhong groups was same to Yuhonggao. The wound surface and healing time were observed and recorded, the diameter of wound surface and count healing rate was calculated, and then draw the wound skin. The sample was handled and embed with Paraffin, HE stained to observe the skin regrowth state and the fibroblast under optical microscope. Trichrome masson stains were performed to watch the array of collagen fibers after wound.And the collagen fibers Area in wound center were counted by Image-Pro-Plus image analysis software.The Substance P expression was measured and compared by immunohistochemistry stain,and the area of SP expression was analyzed by Image-Pro-Plus image analysis software. The nerve fibers were visualized by Holmes silver method and counted.Then the data of SP expression and collagen was dealed by SPSS13.0 statistics software,as well as the numbers of fibroblast,SP positive nerve fibers ,nerve fibers inHolmes. All counted data was expressed in the form of (?)±s, analysis of variance was used to interblock count data.Results:①Average wound healing time hadn't significant difference between the Yuhonggao group and Jingwanhong group, which were better than model group (P<0.05) .②The number of fibroblast in the wound center of Yuhonggao group was more than model group in the 3th, 5th, 7th, 14th day after wound (P<0.05) .There were no difference between the Yuhonggao group and Jingwanhong group about the number of fibroblast in the wound center .③The collagen fibers area in wound center of Yuhonggao group and Jingwanhong group were greater than model group in all days(P<0.05) ,and their collagen was more regularly arranged than model group's in the 21th,30th day after wound.④The SP secretion of Yuhonggao group was more than model group in the 3th,5th,7th,10th,14th day(3,5,7,P<0.01,10,14,P<0.05) ; The SP secretion of Yuhonggao group was higher than Jingwanhong group's in the 3th,5th,7th,10th,14th day (3,7d, P<0.04; 5,10,14d, P<0.05) .The SP secretion of model group was higher than Yuhonggao group and Jingwanhong group's in the 30th day(P<0.05) .The SP positive nerve fibers numbers of Yuhonggao group were more than model group(P<0.05) ,they were often distributed in deep dermis,and near the blood vessels.⑤All the three group's nerve fibers growed gradually increased in the 5th,7th,10th day. Yuhonggao group achieved peak at the the 14th day ,and the others achieved peak in the 21th day.The nerve fibers of Yuhonggao group were more than model group in the 5th,7th,10th,14th,30th day(P<0.05) ,and there were no difference in 3th,21th day.Conclusion: Yuhonggao can accelerate skin wound healing and improve the quality of wound healing. The mechanism of promoting wound healing possibly relate with promoting the Secretion of Substance P in the proliferation period and promoting the reinnervation in the process of rat's skin wound healing.
