Objective: In order to observe the effect of Duxiao Ganqing Keli which fluences liver function consisting ofALT, AST, TBIL, ALB, BA, EEG, LPS, IL—6, TNF-a in the animal model of experimental hepatic encephalopathy in rats and effect of Duxiao Ganqing Keli to gastrointestinal function, furtherly to discuss the therapeutic effect and mechanism in HE.Methods: 1 .Dividatur the rats into 6 groups randamly: blank array, model array, Lactitol array, the diminutive,midst,large dose of Duxiao Ganqing Keli.2.Methods To induce the hepatic encephalopathy the rats were given intraperitoneal injection with. TAA.3.After modeling success,the drugs Duxiao Ganqing Keli.were given to the latter three groups and Lactitol .we given to positive control group until 2 weeks.The normal group was not any treatment and the model group through stomach feeding.by physiological saline.4.After 2 weeks, determine the electroencephalogram.of all the experimental rats. electroencephalographic observation in hepatic encephalopathy. We observe the effects of Duxiao Ganqing Keli treatment of HE on experimental EEG in rats.5.Execute the rats in full after determining the electroencephalogram, abdominal aorta blood was drawn, we test liver function and the serum Ammonia concentration. Observe the contents of LPS, IL—6, TNF-a in hypothalamus of the hepatic encephalopathy rats model with enzyme linked immunosorbent assay (Elisa) We observe the effects of LPS, IL-6, TNF-a by Duxiao Ganqing Keli treatment of HE on experimental EEG in rats.6.We applied electron microscope to observe the developing changes of liver pathobiology of the HE rats model.7. Observe the effect of carbon particle propelling detection to observe the propellent function of small intestine in mice after Duxiao Ganqing Keli's treatment.Results: 1. The effect on the weight, behavioristicsa of HE rats model after Duxiao Ganqing Keli's treatment: Therapeutic groups rats weight gain and nerves reflex enhance, the midst, large dose of Duxiao Ganqing Keli were statistically significant, the lactitol array, the diminutive one were meanningful than than the model array's (P<0.01). the large one was with significant difference in hepatic encephalopathy stages change,and there isn't significant difference in the diminutive one with Lactitol array.2. The effect on the liver function , LPS, and the serum Ammonia of HE rats model after Duxiao Ganqing Keli's treatment: All the indexes decreased notably after treatment to the therapeutic groups than the model array's ( P<0.05),and liver function , BA , LPS decreased notably in the midst,large dose of Duxiao Ganqing Keli, there were significant differences than the model array's (P<0.01). It showed certain improvement after Duxiao Ganqing Keli's treatment.3. The effect on the IL—6, TNF-a of HE rats model after Duxiao Ganqing Keli's treatment: The indexes those were IL—6, TNF-a decreased notably after treatment to the therapeutic groups than the model array's ( P<0.05),and IL—6, TNF-a decreased notably in the midst,large dose of Duxiao Ganqing Keli, there were significant differences than the model array's (P<0.01), the diminutive one decreased too, But it was no difference in the control group.lt showed certain improvement after Duxiao Ganqing Keli's treatment.4. The effect on the electroencephalogram of HE rats model after Duxiao Ganqing Keli's treatment: It was found that the improvement of electroencephalograph of experimental hepatic encephalopathy in rats, compared the model group. They can prevent hepatic encephalopathy and there have different curative effect in different doses, The large group rats' EEG obviously surpass positive control group.5. The effect on the liver pathobiology of HE rats model after Duxiao Ganqing Keli's treatment: The hepatocell degeneration and necrosis and delaying the formation of pseudolobe in the positive control group,the diminutive,midst,large dose ofDuxiao Ganqing Keli. It indicate that Duxiao Ganqing Keli can reduce the hepatic injury and protect the liver when the liver is injured in HE rats.6. The effect on the peristalsis experimental rats model after Duxiao Ganqing Keli's treatment: The treatment groups are better than the contrast group in the carbon powder's advance percentage (P<0.01, P<0.05). The midst,large dose of Duxiao Ganqing Keli have remarkable significance in contrast with positive group . There is preferable effect of intestinal peristalsis to Duxiao Ganqing KeliConclusion:1. We can produce the hepatic encephalopathy model of rats successfully by the rats were given intraperitoneal injection .with TAA.2.The consequence to the research manifest that: The consequence to the research manifest that: Duxiao Ganqing Keli can improve hepatic function,BA,LPS, IL—6,TNF-a,liver pathobiology,electroencephalogram of the HE rats in a certain extent, the midst,large dose ofDuxiao Ganqing Keli is efficient. On the whole, it is effective than the positive control group.3. Duxiao Ganqing Keli is a sort of active drug used to prevention and cure the hepatic encephalopathy, and is worthy of further study and development.
