The Chinese Traditional Medicinal Compound Recipe (CTMCR) Gastrodine pellets were composed of Rhizoma Gastrodiae, Fructus Schizandrae and Ophiopogonis Tuber. The Rhizoma Gastrodiae in this recipe was used to calm and ease pain. Schisandra Chinensis and Ophiopogon Japonicus were assisted. While the three were combined, the effects of strengthening and invigorating the body, and treating both symptom and radical were exerted. The recipe was shown to be effective in insomnia, forgettery, neurasthenic and in dizziness and headache caused by hypertension. Good curative effect and low side effect of the marketed granules were gained. Based on extraction and purification of Rhizoma Gastrodiae, Schisandra Chinensis and Ophiopogon Japonicus, the pellets were prepared to reduce the dose of administration and would be more convenient for patients.In this research, Rhizoma Gastrodiae, Schisandra Chinensis and Ophiopogon Japonicus were identified by TLC. The method was established for determining the content of Gastrodine and Deoxyschizandrin by HPLC, the linear range of Gastrodine was 10~80mg·L-1. The precision, stability and recovery were excellent. The methods of content determination of Rhizoma Gastrodiae and Schisandra Chinensis were both exact, reliable and had good reproducibility.Orthogonal experiment design and single factor tests were used in optimizing the extraction and purification technology of Rhizoma Gastrodiae measured by Gastrodine Transfer Rate. Gastrodine was accumulated by using macroporous resin D101 as filler. Supercrtical Fluid Extraction (SFE) was used to extract Deoxyschizandrin. By the single factor tests measured by extraction yield of Deoxyschizandrin, the technical conditions were determined: materials input 300g, extraction pressure 30 MPa, extraction temperature 50℃, extraction time 120min. Such factors as material crush, the quantity of water, extraction time and extraction frequency were investigated in the decocting technology. These factors were measured by output rate and leaching rate. The optimized technical condition showed that the extract yield was high and the repeatability was good.Gastrodine pellets were prepared by extrusion-spheronization. The powder parameters and yields were investigated. Through the formulation screening and process observation, it was showed that the yield of the objective pellets was above 80%. The pellets prepared by this method had perfect shape and surface characteristics.The formulations stability studies showed that the Gastrodine pellets were stable when exposed to light, moisture and heat. The results of accelerated testing in six months showed that there was no much change in the invested parameters, and the preparation stability was satisfied.The pharmacodynamic effects of relieving fever, consciousness regaining and easing pain were studied. The results showed that CTMCR Gastrodine pellets exhibited an obvious effect of relieving fever, which was not clearly influenced by dosage. Gastrodine pellets increased evidently the times of mouse's conscious activities, prolonged latent period to fall asleep and shortened hours of sleep both caused by pentobarbital sodium, extended markedly stretching latent period and lessened stretching times in 15 min caused by acetic acid. Taking one with another, the pharmacodynamic actions of CTMCR Gastrodine pellets were equivalent to those of aspirin, without prominent difference.
