Higf-i And Of Hil-1¦Â On The Role Of The Degeneration Of The Cartilage Endplate Cells

Object: To approach the effect of hIGF-I on the intervertebral discsendplate cell culture in vitro of New Zealand white rabbit. To observe the effect of hIGF-I on endplate cell, with and without hIL-1βas the intfc factor. To study and research the dose-effect relationship between these two growth factors on rabbit intervertebral discs endplate cell culture in vitro.Methods: To separate homology young New Zealand rabbit cartilageendplate cell and culture it in vitro. To obtain degenerated endplate cell with the principle of mature animal normal rhagiocrine cell degeneration with high passage. Different dose of hIGF-I and hIL-1βhade been added on endplate cell and the effect of these two factors on cell excretion of collagen-Ⅱand glucoprotein want to be observed with immunohistochemistry methods. Research and study the data collected from these tests with statistical method, and then calculate the dose-effect relationship curves of hIGF-I and hIL-1βon endplate cell.Result: hIGF-I has obviously antagonism effect to hIL-1βon differentgenerations of endplate cell. hIGF-I can promote the protein excretion of collagen-Ⅱand glucoprotein on these generations of the cell. The dose-effect relationship curves between hIGF-I, IL-1βand endplate cell were also been calculated.Conclusion: hIGF-I can promote the protein excretion of collagen-Ⅱand glucoprotein under the environment with and without the inflammatory factor IL-1β. hIGF-I also can oppose the repress effect of IL-1βon endplate cell. The effect of hIGF-I and hIL-1βon endplate cell is under dose-depended relationship which can be described with regression equations.
