Aim: To discover neurotrophic or(and) nerve injury protective materials from medicinal resource of Guizhou Province by large scale screening, four screening models will be developed and optimized based on PC12 cells in this work, and these models will be evaluated by screening some plant extracts or compounds which originally came from medicinal plants used in ethnodrugs of Guizhou.Method: Rat pheochromocytoma cells( PC12 cells )possesses character of stabilization and homogencity, and can respond to NGF rapidly. PC12 cells usually were used in various studies related to nerve cell differentiation and function. In this work PC12 cells were employed to establish four screening models for discovery of neurotrophic and nerve injury protective materials. These four models involved nerve cell differentiation and injury induced by hydrogen peroxide,glutamate andβ-amyloid pepide, respectively, were duplicated. The models were optimized according to the assays of some major conditons in the models such as cell density, the concentration of injury reagents and the time exposed to injury reagents. After the cytotoxity of samples was assayed by MTT method, the working concentrations of samples were determined. Then the neurotrophic and (or) nerve injury protective activities of these samples were evaluated by the models mentioned above.Results: Four models involved nerve cell differentiation and injury induced by hydrogen peroxide,glutamate andβ-amyloid pepide, respectively, were duplicated and optimized successfully. More than 588 samples were screened by using those models, and some samples were found to possess neurotrophic or (and) nerve injury protective activities.Conclusion: The four models constituted in this work are practicable, effective and reliable. These models are suitable for screening neurotrophic and (or) nerve injury protective materials from medicinal resource of Guizhou ethnodrug. Screening practice in this work can provide some guide to large scale screening in next step.
