Objective: To investigate the chromosome and gene alterations in adenocarcinoma of lung cell line, OM.Method: Cytogenetic and molecular methods, such as G-banding, multi-color FISH, CGH (comparative genomic hybridization), and PCR were carried out to analyze chromosome changes and gene alteration using adenocarcinoma of lung cell line,OM.Results:1. G-banding showed chromosomal aberrations on all chromosomes except, 4, 8, 14, 15, 17, and 19.2. Multi-color FISH revealed the chromosome translocations and origin of marker chromosomes: Four translocations on 10.Two translocations on 2,5,9,11,18. One translocation on 3 and 21.3. CGH revealed the gains on chromosome regions of 3q24-28, 5p13, 12q22-23.24 and the losses on 3p24.1-26, 6p25, 6q25.2-27, 7q32-36, 8p22-23, 9p21-24, 10q25-26.3, 12p13.31-13.33,17p13.1-13.3.4. PCR showed the amplification of genes: MME, SI,BCHE,SLC2A2 and KNG on 3q.Conclusion:1. Complex karotypes were seen on the adenocarcinoma of lung cell line, OM, by chromosome analysis.2. Tanslocation involved chromosome 10 may play an important role in the adenocarcinoma of lung cell line, OM.3. The existence of oncogenes on the 3 chromosomes and tumor suppressor genes on 8 chromosomes were suggested by CGH analysis.4. In PCR analysis, MME,SI,BCHE,SLC2A2 ,and KNG genes on chromosomes, 3q may play a pivotal role in carcinogenesis on OM lung cancer cells.
