Expression of Neuroglobin mRNA in brain tissue of rats after cerebral ischemiaObjective: To study the expression of Ngb mRNA after rat cerebral ischemia and to find out thevalue of Ngb mRNA in estimating the trauma time in forensic medicine aspect.Methods: Set up a Wistar rat model of cerebral ischemia by clamping both arteria carotiscommunis. Wistar rats were killed at 0min,1min,5min,10min,30min,60min after respectivelycerebral ischemia interval. Total RNA isolated was evaluated, and then, the non-degradationRNA was carried out by one step RT-PCR. GAPDH was used to be inside reference. Then NgbmRNA and GAPDH mRNA were amplified by specifical primer. After 1.5% agarose gelelectrophoresis, the ratio of Ngb mRNA/GAPDH mRNA was detected by gel image analysistechnology and analyzed the correlation to different cerebral ischemia interval.Results: (1) The expression of Ngb mRNA after ischemia trauma was definite and generous.(2) The expression of Ngb mRNA rose quickly after cerebral ischemia, revealed that Ngb was sensitive to hypoxia and maybe protected neuron.(3) The expression of Ngb mRNA correlated to cerebral ischemia interval. It reached peak value after 1min, obviously different to control group(P<0.01); After 5min it decreased until 10min, nonsignificance compared to control group(P>0.05); After 10min it rose gradually again according to ischemia time, obviously different to control group(P<0.01) too.Conclusion: The expression of NGB mRNA in cerebral tissue increased rapidly after ischemia, suggesting that NGB may have important function in the adaptative protection process of hypoxic-ischemic brain damage. The expression of Ngb mRNA changed according to different time after cerebral ischemia, and it may be used to estimate the early time of ischemia trauma in forensic practice.2. Study on sex estimation by fibulaObjective: To investigate the method of sex estimation by fibula and to establish the sex discriminant functions by the parameters of fibula.Methods: 610 fibulas were chosen from 305 known Chinese skeletons (274 males and 31 females, age ranges from 18 to 84), which come from 9 provinces of China. Through anthropometric methods ,5 parameters were measured with measurement instruments. Data obtained were analyzed by Statistical Package for Social Sciences 11.5 (SPSS 11.5). Independent-samples t test was made between left and right of fibula first, and then between male and female. Data were subjected to Fisher discriminant analasis, and parameters that showed difference significantly were selected for constructing the sex discriminant functions.Results: (1) The fibulas of male or female can not be differ from each other by sides, we can choose the right fibulas to continue our study.(2) All of the 5 parameters can be used for sex estimation.(3) 5 single factor discriminant functions and 1 multiple factor discriminant function were obtained. The accuracy of the 5 single factor discriminant functions is between 61.64% and 75.74%, and the accuracy of the multiple factor discriminant function is 82.62%.Conclution: It showed that the discriminant functions acquired were effective in the sex determination for Chinese, and the multiple factor discriminant function is more reliable and valuable. |
