Guogong medicinal wine, with its efficacy of Sanfengqushi and Shujinhuoluo, is widely used in clinical applications. Due to historical reasons, the quality control of product should be further enhanced, and manufacturing process could be further enhanced. According to the above problems, Guogong medicinal wine was selected to develop quality evaluation methods, on-line detection, and process comparison. Main content and the results are as follows:1. The paper developed some methods to determine hesperidin, naringin, neohesperidin, ferulic acid, isoimperatorin, isoimperatorin, osthole, honokiol, neohesperidin and magnolol in Guogong medicinal wine by HPLC. Ferulic acid, isoimperatorin, isoimperatorin, osthole and neohesperidin were first determined in it. The results show that the methods were proved to be accurate, reliable, and they can be used to control quality of Guogong medicinal wine. In addition, 10 batches of samples were determined by the methods, and content ranges of 9 compounds were found preliminary.2. The fingerprints of Guogong medicinal wine were investigated by HPLC. The method was validated and 21 batches of samples were investigated. Software of fingerprint evaluation similarity was used to evaluate similarity of fingerprints among different samples. And the similarity of between samples and comparison fingerprint is greater than 0.97. 42 common peaks were found,and 9 compounds were identified by comparing retention time, adding reference substances and employing DAD technology.3. The fingerprints of Guogong medicinal wine were investigated by GC-MS. The method was validated, and the outcome show that it can be used to control volatile components in Guogong medicinal wine. 24 batches of samples were investigated, and the similarity between the comparison fingerprint and fingerprints of samples is greater than 0.9, except No.3180060 and No.4180004. The structures of 26 common peaks in fingerprint were identified by mass spectra.4. 22 batches of samples were determined by UV spectroscopy and HPLC. Correlation analysis was used in results determined by UV spectroscopy and HPLC. And an UV model for predicting the content of hesperidin was established. The predicting equation was Chesperidin (ug·mL-1) =347.4×A284nm+56.29 (r=0.9160, n = 22).By measuring the absorbance at 284nm, the content of hesperidin can be predicted. As the absorbance of brown sugar in UV spectra is large, the aqueous solution of brown sugar (diluted 20 times) was selected as a reference solution. To eliminate interference of different batches of brown sugar in UV spectra, the reference solution must be changed when brown sugar which was used in manufacture changed. Another 11 batches of samples were used to validate the predicting model, and the predicting results are good.5. Correlation analysis was used in results determined by HPLC and NIR spectroscopy. First order derivative spectra were selected, and SMLR was used to data processing. The wave numbers of 4589.75,7721.58,8296.26 and 9214.21 cm-1were selected to build the model for predicting the content of hesperidin in Gouging medicinal wine. The predicting equation was Chesperidin(ug·mL-1)=3.334790×103X1-1.676656×106X2-2.293503×106X3+2.157171×106X4+4.428600×102(r=0.94002, n=23). X1, X2, X3, X4 were values of first derivative of 4589.75, 7721.58, 8296.26 and 9214.21cm-1. Another 10 batches of samples were determined to validate the predicting model, and the predicting results are good.6. Content of 9 compounds, HPLC fingerprints and GC-MS fingerprints of samples in the settlement period and samples before and after ultra filtration were investigated. The similarity of fingerprints in settlement period is greater than 0.98. The content of 9 compounds in settlement period changed little. The similarity of fingerprints before and after ultrafiltration is greater than 0.99, and the content of 9 compounds have no significant change. The results provide the data to shorten the settlement period by using ultrafiltration.
