Object To study the inhibitory effect of vincristine(VCR) on human squamous cell carcinoma (Ta-8113) anaplastic and the synergic anti-tumour effect of verapamil(VPL) with vincristine.Methods Tca-8113 were exposed to vincristine respectively, and to verapamil combined with vincristine at various concentrations. The inhibitory effect and synergic anti-tumor effect were detected by microculture tetrazolium colorimetric(MTT) assay. Every group included three holes. The cells of group A were cultured with no drugs as blank. The cells of group B were cultured with vincristine (seven kinds of concentration) as control. Group C was with vincristine(seven kinds of concentration) and verapamil(5ug/ml). Group D was with vincristine (seven kinds of concentration) and verapamil(10ug/ml). Group E was with vincristine(seven kinds of concentration) and verapamil(20ug/ml).The absorbance of every group was detected by MTT. Results1. The survival rate of every group decreased when the concentration of vincristine increased.2. The survival rate decreased when the concentration of verapamil increased at the same concentration of vincristine.But the survival rate decreased rarely between 10ug/ml and 20ug/ml of verapamil.3. Between 0.05ug/ml and 100ug/ml of vincristine, the survival rate of group B decreased with the increase of vincristine's concentration.4. Between 0.05ug/ml and 100ug/ml of vincristine, the survival rate of group C was less than 30%. It suggested that the cells were hyper sensitive about drug combination. The survival rate of group D was less than 30%. It suggested that the cells were hyper sensitive about drug combination. The survival rate of group D was less than 30%.It suggested...
