| Background:The topic is a branch of the study of a kind of nonspecific antidotic Chinese crude drugs, its new ideas are based on " nonspecific deintoxication". Concerning the application of antidote at present, there are several regrets: 1.The deficit of specific antidote; 2.Great part of antidotes are specific, the spectrum of diseases they overjut is very narrow;3.The safety margin of some antidotes is narrow also. Otherwise, it is difficult for hospitals to give a precise diacrisis to the toxicopathy quickly and punctually. Respecting of the state mentioned above, it is very necessary to research a kind of nonspecific antidote. JDF has been proved a very efficacious prescription to many kinds of toxicopathy for more than thirty years by my advisor. Accordingly, studying its mechanism and target of action is very significant. Endotoxemia is a symptom complex that is caused by Gram-negative bacteria`s endotoxin entering blood, whose key feature is febricity(T≥38.5℃).Endotoxemia belongs to toxicopathy. Endotoxemia can cause systemic inflammatory response syndrome (SIRS),multiple organ dysfunction syndrome(MODS), even multiple organ failure(MOF). Therefore, basical and clinical researchs of endotoxemia are very necessary so as to avoiding serious consequence.Methods:40 Wistar rats were randomly and equally divided into 4 group:Group of NS, LPS, LPS+prescription, LPS+DEX. The animals in every group were intragastric administrated for 4 days in advance, 2 times a day (Group of LPS+prescription: equivalent traditional Chinese drugs, other three groups: deionized water of equal volume).All animals were intraperitoneal injected continual(30mg/kg) two hours after the first intragastric administration in the fifth day (at the same time, the animals in the Group of LPS+DEX were intraperitoneally injected DEX 5mg/kg). After the animals were anesthetized, vena caudalis injection started(Group of NS: NS of equal volume, the other three groups: LPS4mg/kg).All animals were intragastric administr- ated 2 hours,6 hours,10 hours after vena caudalis injection(The group of LPS+prescr- iption: 2 times equivalent traditional Chinese drugs this day).24 hours after the vena caudalis injection, all rats were killed for the materials. Firstly, observe the animals` spi- rity, activity, fur, weight etc. Secondly, measure the blood serum BUN, Cr, ALT, AST, CK, CKMB. Thirdly, observe the pathological sections of heart, lung, liver and kidney.Results:1. Activity: Group of NS: acted and sed normally; Group of LPS: less acting, depressed, piloerection, heaping; Group of LPS+prescription: acting well, same as the Group of NS; Group of LPS+DEX: active, stimulated.2. Weight: comparing with the weight at the time of vena caudalis injection, the animal`s weights in the group of NS recovered nearly; the animals` weights in group of LPS decreased obviously(P<0.01);JDF can improve this tendency(P<0.01);DEX can aggravate this tendency(P<0.01).3. BUN,Cr:comparing with the group of NS, animals` blood serum BUN, Cr in group of LPS increased obviously(P<0.01); JDF can degrade Cr`s increasing(P<0.01); DEX can promote BUN`s increasing ( P<0.01 ) ;otherwise, comparing with the group of LPS+prescription, animal`s blood serum Cr in group of LPS+DEX increased(P<0.01).4. ALT,AST:comparing with the group of NS, animal`s blood serum ALT in group of LPS decreased obviously(P<0.01); JDF caould improve this tendency(P<0.01).5. CK,CKMB:comparing with the group of NS, animal`s blood serum both CK and CKMB in group of LPS decreased obviously(P<0.01);DEX can improve this tendency(P<0.01),this role of JDF is not conspicuous.6. Pathological section: there is no significant deviation during 4 groups on lung; JDF can improve the injury of heart, liver and kidney caused by LPS, whose defend of kidney is conspicuous even, its defend of liver is less than DEX; DEX can guard heart and liver, however, it may aggravate kidney injury, mainly on renal tubule injury.Conclusions:1. Both JDF and DEX can improve the rats`s depressed spirity and activity caused by LPS, the rats may be unduly stimulated for DEX injection.2. JDF can improve the rats`s weight decreasing caused by LPS, however, DEX may aggravate this tendency.3. JDF can defend the kidney injury caused by LPS, however,DEX may aggravate this tendency.4. Both JDF and DEX can defend liver injury caused by LPS, the latter may surpass the former.5. Both JDF and DEX can defend myocardial damage caused by LPS, the latter may surpass the former... |
