Diabetes And Its Complications In The Treatment Of Drug Screening

Purpose:The medicines with having the effect of treating diabetes and the complication were screened out by three diabetic models in vitro. And the curative effect of traditional Chinese medicines compounded on diabetes mellitus and the complication were observed. Methods:On the basis of diabetic biochemistry mechanism, we screen out many medicines by the inhibitor of aldehyde carbohydrate deoxidize enryme system, the radicals scavenger system and the inhibitor of glycosylated protein in vitro, and the hypoglycemic effect of the medicines on the diabetes in the mice induced by Alloxan monohydrate in vivo. Finally ,we comfirmed the Berberis Poiretii Schneid and Suberect spatholobus stem as the components of the compounded medicines and optimized proportion in vitro. The treatment effect of BSS on diabetes mellitus and the complication was observed in the diabetic rats induced by streptozotocin. The type symptoms of diabetes mellitus in the diabetic rats, such as the body weight, the drinking water volume, the urine volume etc were observed. The changes of the tissue organs structure and function in the diabetic rats, such as heart, kidney, lens, nerve etc were examined. And the various biochemistry indexes in the blood and related with diabetes mellitus and the complication were detected. Results:Medicine screening : Suberect spatholobus stem, Corydalis tuber, Chinese corktree bark and Chinese angelica etc have rather strong inhibition effect on the production of Sorbitol in the incubated red blood cell in vitro. The extracts from Guava leaf, Chinese nut-gall, Dwarf lilyturf tuber, Suberect spatholobus stem etc can inhibit the production of malondialdehyde (MDA) in the liver well-distribution slurry in vitro. The extracts from Berberis Poiretii Schneid, Chinese angelica, Dwarf lilyturf tuber and Suberect spatholobus stem can inhibit the production of Glycated protein in the incubated liquid in vitro. Berberis Poiretii Schneid has the obvious hypoglycemic effect on the diabetic mice induced by Alloxan monohydrate, however, Suberect spatholobus stem has not hypoglycemic effect but the conditions of mice get the improvement.Pharmacodynamics experiment:In the model rats induced by streptozotocin, rats body weight obviously decreased, drinking water volume and urine volume apparently increased, death rate rised; blood sugar, Glycated serum protein, Glycated ferrohemoglobin, Sorbitol in red blood cell strikingly increased; the content of MDA in the brain obviously increased; CK, LDH in the plasma apparently increased; the content of plasma carbamide nitrogen and urine protein obviously increase; the latent period of sense of pain extended, nerve conduction velocity of rat tail strikingly slowed down., the content of Sorbitol in the ischium nerve obviously increased, instead of Cyclohexanehexol depressed; the heart,kidney,pancreas,ischium nerve, etc, had all had pathological changes at the different extent. The results indicated that BSS (150,300,600mg/kg ig.)improved the decrease of rats body weight, the increase of drinking water volume and urine volume and death rate, reduced strikingly the blood sugar, Glycated protein, Glycated ferrohemoglobin, and Sorbitol in red blood cell; decreased obviously the content of MDA in the brain and the CK,LDH in the plasma, and the increasing of plasma carbamide nitrogen and urine protein alse obviously were ameliorated; the latent period of sense of pain was shorten, amended nerve conduction velocity of rat tail, reduced the content of Sorbitol in the ischium nerve. BSS can improve these pathological changes on the heart,kidney,pancreas,and ischium nerve etc. In conclusion,BSS had certain treatment effect on the diabetes mellitus and the complication in the rats.Conclusion: Many extracts of traditional Chinese medicines have rather strong inhibitive effects on the production of Sorbitol, radicals, and Glycated protein in vitro. The BSS screening by vitro experiments have the certain treatment effect on the diabetes mellitus and the complication.
