Compound Tramadol Hydrochloride Tablets

Tramadol hydrochloride was widely used to treat acute and chronic pain as its low side effect. Recently, there were a number of clinical research which show the combination of tramadol and diazepam .These research show that the two drug provide synergism of easing pain and antagonism of toxicity. Some research about the dose-efficacy relationship and drug efficacy were done on the basis of clinical research in this paper. Compound tramadol tablet and two-layer sustained release tablet was designed and prepared.The dose-efficacy relationship of tramadol and diazepam was studied with hot-plate test on mouse. The best compatibility of medicines was filtered. The drug efficacy was proved with hot-plate test on mouse and pressure pain test on rice. The Q value werel.57and 2.09.The result indicated that the two drug provided significant synergism. The ED50 were 11.67mg/kg and 7.09mg/kg. The acute toxicity was investigated with Kou' s test.According to the literature, two-wavelength method was developed for the determination of physiochemical properties and drug release. High performance liquid chromatography (HPLC) method with UV detection was applied to the determination of content, examination of impurity and the plasma concentration of tramadol and diazepam in dogs.In the preformulation research of compound tablet, orthogonal experiment was used to optimize the formulation.
